超临界萃取海参脑苷脂及HPLC检测分析

中文摘要: 通过超临界萃取的方式提取海参中的脑苷脂,利用单因素试验研究萃取温度、萃取时间、萃取压力以及海参粉的颗粒大小对超临界萃取海参中的脑苷脂提取率的影响,用正交试验来优化超临界萃取的条件,使用高效液相色谱(HPLC)来检测海参中脑苷脂的提取率。试验结果表明,适宜的超临界萃取条件是:萃取时间3 h,萃取温度38℃,萃取压力20 MPa,颗粒物大小60目,用高效液相色谱法分别测得了半乳糖脑苷脂的提取率,在前述测定条件下,海参中的脑苷脂最高提取率可达0.107 9%。
Abstract:By supercritical fluid extraction to extract cerebroside from sea cucumber,and through the single factor experiment to explore the extraction temperature,extraction time,extraction pressure and sea cucumber powder particle size on the cerebroside extraction yield of supercritical fluid extraction,using the orthogonal experiment to optimize the supercritical fluid extraction conditions,using high performance liquid chromatography(HPLC)to detect the sea cucumber cerebroside extraction yield.The experimental results showed that appropriate conditions of supercritical fluid extraction was extraction time 3 h,extraction temperature 38℃,extraction pressure 20 MPa,the particle size 60 mesh.The extraction yield of galactose cerebroside was measured by HPLC method.Under the determination conditions above,the highest extraction yield of cerebroside in sea cucumber was 0.107 9%.
