中文摘要: 研究超声辅助双水相提取菠萝皮总黄酮和多糖的最佳工艺条件。以萃取率为指标,采用超声辅助磷酸氢二钾-乙醇双水相同时提取菠萝皮总黄酮和多糖,通过单因素试验和正交试验考察料液比、温度、超声时间、乙醇溶液浓度和磷酸氢二钾溶液浓度对提取的影响。结果表明黄酮最佳提取工艺为乙醇体积分数95%,料液比1∶30(g/mL),磷酸氢二钾质量分数为30%,提取温度50℃,超声时间20 min。多糖最优提取工艺为乙醇体积分数70%,料液比1∶45(g/mL),磷酸氢二钾质量分数55%,提取温度60℃,超声时间20 min。在最优工艺条件下经过3次平行试验,提取3次,黄酮和多糖萃取率分别为88.64%、86.91%,得率分别为21.33、18.95 mg/g。说明工艺条件稳定可行,结果重现性较好。

Abstract:The optimum extraction process of flavones and polysaccharides from pineapple peel were studied by ultrasonic-assisted aqueous two-phase system.The aqueous two-phase were mixed by dipotassium phosphate solution and ethanol solution.It had been researched that solid-liquid rate,the temperature,the ultrasonic time,ethanol solution concentration and dipotassium phosphate solution concentration had effect on extraction rate by means of single factor and orthogonal experiment method,which used extracts rate as the evaluation standard.The results showed that the optimal extraction conditions of the flavones were ethanol solution concentration 95%(volume fraction),solid-liquid rate 1 ∶30(g/mL),dipotassium phosphate solution concentration 30%(mass fraction),the temperature 50℃ and the ultrasonic time 20 minutes,and the optimal extraction conditions of the polysaccharides were ethanol solution concentration 70%(volume fraction),solidliquid rate1∶45(g/mL),dipotassium phosphate solution concentration 55%,the temperature 60℃ and the ultrasonic time 20 minutes.After three parallel experiments and three times extraction the average extraction rate of the flavones and polysaccharides was 88.64%and 86.91%,and the yield was 21.33 mg/g and 18.95 mg/g,respectively.The results were stable and feasible.
