中文摘要: 为优化藏羊骨肽的制备工艺条件以及评估其体外抗氧化和降血脂活性,以藏羊骨为原料制备骨肽,以酶解液的水解度作为评价指标,在筛选适宜蛋白酶基础上,通过单因素试验确定适宜蛋白酶的pH 值、温度、底物浓度、酶添加量和酶解时间范围,进一步通过响应面试验确定最佳酶解条件。以DPPH 自由基清除率和ABTS+自由基清除率为体外抗氧化指标,以胰脂肪酶抑制率为体外降血脂指标,评估骨肽的抗氧化和降血脂能力。结果显示,水解藏羊骨肽的最适酶为碱性蛋白酶,最佳酶解条件为pH10.3、温度45 ℃、底物浓度为10%、酶添加量8%、酶解时间6 h,在此条件下,水解度可达到35.13%。藏羊骨肽的DPPH 自由基清除率为69.97%,ABTS+自由基清除率为81.25%,其IC50 值分别为(3.78±0.12) mg/mL 和(2.72±0.06) mg/mL。骨肽对胰脂肪酶的抑制率为43.59%,IC50 值为(14.48±0.51)mg/mL。该研究确定了藏羊骨肽的最佳酶解条件,并初步表明其具有较好的体外抗氧化和降血脂活性。
Abstract:To optimize the preparation conditions of Tibetan sheep bone peptide and evaluate its antioxidant and hypolipidemic activities in vitro,peptides were prepared from Tibetan sheep bone. The degree of hydrolysis of the enzymolysis solution was used as the evaluation index. Suitable proteases were screened,and singlefactor tests were used to determine the optimal pH,temperature,substrate concentration,enzyme amount,and enzymolysis time for the selected proteases. The optimal enzymolysis conditions were further refined using a response surface test. In vitro antioxidant activity of the peptides was assessed by using the DPPH free radical scavenging rate and the ABTS+ free radical scavenging rate,while the inhibition rate on pancreatic lipase served as an indicator of hypolipidemic ability in vitro. The results indicated that the most suitable enzyme for hydrolyzing Tibetan sheep bone peptides was alkaline protease. The optimal enzymolysis conditions were as follows:pH of 3,temperature of 45 ℃,substrate concentration of 10%,enzyme amount of 8%,and enzymolysis time of 6 h. Under these conditions,the degree of hydrolysis reached 35.13%. The scavenging rates of Tibetan sheep bone peptides for DPPH and ABTS+ free radicals were 69.97% and 81.25%,respectively,with IC50 values of (3.78±0.12) mg/mL and (2.72±0.06) mg/mL,respectively. The inhibitory rate of bone peptides on pancreatic lipase was 43.59%,with an IC50 value of (14.48±0.51) mg/mL. This study successfully determined the optimal enzymolysis conditions for Tibetan sheep bone peptides,and preliminary results indicated that these peptides exhibited significant antioxidant and hypolipidemic activities in vitro.
