中文摘要: 该文研制可用于肉及肉制品中海狸鼠源性成分定性的标准样品。采用经验证确认的海狸鼠肌肉组织作为原料,经粉碎混匀并分装,包装并辐照后制成标准样品。对样品进行均匀性和稳定性检验,并对样品进行定值,获得样品的特性值。结果表明,样品原料采用实时荧光聚合酶链式反应(polymerase chain reaction,PCR)方法和测序方法验证结果均为海狸鼠源性成分阳性,且无其他源性成分检出;样品的均匀性检验和短期稳定性检验结果均为海狸鼠源性成分阳性,Ct 值经方差分析表明在95%置信水平下单元间不具有统计显著性,证明样品是均匀的,在模拟运输条件下稳定性良好;经趋势分析,结果表明样品在24 个月内稳定性良好;样品开封后检验结果均为海狸鼠源性成分阳性,Ct 值检验结果经方差分析在95% 置信水平下不具有统计显著性,说明样品在-18 ℃条件下90 d 内反复冻融7 次依然稳定;样品经10 家实验室联合定值,检测结果均为海狸鼠源性成分阳性,阳性率为100%。样品均匀性、稳定性和赋值均满足标准样品的要求,可用于相关检测方法的证实以及检测全程的质量保证等,能够满足标准样品的预期用途。
Abstract:The reference materials for Myocastor coypus-derived components in meat and meat products were developed. The validated muscle tissue of Myocastor coypus was used as raw material, crushed, mixed well,packaged, and irradiated to make reference materials. The homogeneity and stability of the reference material were tested, and the property values of the reference materials were determined. The results showed that the raw materials were confirmed to be positive for Myocastor coypus by polymerase chain reaction (PCR) and sequencing methods, and no other derived components were detected. The homogeneity and the short-term stability test results of the reference materials were positive for Myocastor coypus. The Ct values were analyzed by variance, and the results showed that the element was not statistically significant at the 95% confidence level,indicating that the reference materials were homogeneous, showing good stability under simulated transportation conditions. According to the trend analysis, the reference materials had good stability within 24 months.After the reference materials were opened, the test results were all positive for Myocastor coypus. The Ct values tested by variance showed no statistical significance at the 95% confidence level, indicating that the reference materials remained stable after repeated freeze-thaw seven times within 90 days under -18 ℃ conditions. The reference materials were jointly evaluated by 10 laboratories, and the test results were all positive for Myocastor coypus, with a positive rate of 100%. The homogeneity, stability, and property values of the reference materials fulfilled the certified reference materials. They could be used for the verification of relevant detection methods and the quality assurance of the whole test process, meeting the intended use of reference materials.
