一株抗真菌卡利比克迈耶氏酵母(Meyerozyma caribbica lut-Y1)的鉴定及其抑菌活性

中文摘要: 该文从宁夏枸杞果实霉腐斑中分离得到一株能拮抗多种病原真菌的酵母lut-Y1 菌株,研究该菌株的菌落形态、细胞形态、生长曲线特征及适宜的生长温度、pH 值及盐耐受度,用平板对峙法观察该菌株对链格孢菌(Alternaria alternate)、小孢壳二孢(Ascochyta leptospora)、细极链格孢菌(Alternaria tenuissima)的拮抗效果,并分析该菌株发酵液对3 种病原真菌的抑菌活性。结果表明,酵母lut-Y1 菌株在PDA 液体培养基上生长良好,能分泌红色素,并具有酵母菌的一般形态特征。根据形态、rDNA-ITS 区测序比对和系统进化树分析将酵母lut-Y1 菌株鉴定为卡利比克迈耶氏酵母(Meyerozyma caribbica)。酵母lut-Y1 菌株在PDA 液体培养下,6~18 h 后进入对数生长期,18~36 h 为稳定生长期,其适宜的生长温度和pH 值分别为30 ℃和6~7,最适生长NaCl 浓度为0%~3%。酵母lut-Y1 菌株菌落与3 种霉菌菌落之间形成明显的抑菌带。该菌株发酵液浓度与3 种病原真菌生长圈直径之间均呈极显著负相关(P<0.01),根据拟合方程,发酵液浓度分别为42.04%、38.42%、37.03%时,可完全抑制链格孢菌、小孢壳二孢、细极链格孢菌生长。酵母lut-Y1 菌株可作为一种有广谱抑菌效果的生物防治菌,应用于果蔬防腐。
Abstract:A strain of yeast lut-Y1 that antagonized many pathogenic fungi was isolated from the fruit mold rot of Lycium barbarum in Ningxia. The strains were studied in terms of the colonial and cellular morphology,growth curve characteristics as well as suitable growth temperature, pH and salt tolerance. Furthermore, the antagonism effect of strain lut-Y1 on Alternaria alternate, Ascochyta leptospora and Alternaria tenuissima was observed. The antifungal effect of strain lut-Y1 broth on three pathogenic fungi was analyzed. The result showed that yeast strain lut-Y1 grew well in PDA broth, with secreted red pigment and the general morphological characteristics of yeast. Based on the morphology, rDNA-ITS region sequencing, and phylogenetic tree analysis, the strain lut-Y1 was identified as Meyerozyma caribbica. In the PDA broth, the growth of lut-Y1 strain entered the exponential growth phase after 6-18 h, and the stable growth phase after 18-36 h. The suitable growth temperature and pH were 30 ℃ and 6-7, respectively. The optimum NaCl concentration for growth was 0%-3%. Obvious bacteriostatic bands were formed between the strain lut-Y1 and the three molds. There was a significant negative correlation between the concentration of fermentation broth and the growth circle diameter of the three pathogenic fungi (P<0.01). According to the fitting equation, when the concentration of fermentation broth was 42.04%, 38.42% and 37.03%, the growth of Alternaria alternate, Ascochyta leptospora and Alternaria tenuissima could be completely inhibited, respectively. The yeast strain lut-Y1 could be used in fruit and vegetable preservation as a kind of biocontrol bacteria with a broad-spectrum antifungal effect.
