中文摘要: 为明确真姬菇多糖(Hypsizygus marmoreus polysaccharide,HM-P)的结构组成及体外抗肿瘤活性,采用热水浸提法分离提取HM-P,利用凝胶渗透色谱、傅里叶变换红外光谱和高效液相色谱等技术鉴定HM-P 的组成成分,采用细胞活力检测试剂盒(cell counting kit-8,CCK-8)以及细胞划痕法检验HM-P 的体外抗肿瘤活性。结果表明,HM-P的重均分子量(weight average molecular weight,MW)为25 232 Da,由半乳糖、葡萄糖、木糖以72.406∶17.661∶9.933 的含量比例组成。HM-P 能够显著地抑制肺癌细胞(A549)、结肠癌细胞(CT26.WT)、成纤维细胞(L929)和胃癌细胞(MFC)增殖(P<0.05)、迁移(P<0.05),以及极显著地抑制腹水癌细胞(S180)增殖(P<0.01),其中HM-P 对胃癌细胞(MFC)的直接抑制作用最强,当HM-P 终浓度为20 μg/mL 时,抑制率达到27.9%,迁移率达到19.0%。HM-P 通过介导巨噬细胞(RAW264.7)上清液杀死肺癌细胞(A549)、结肠癌细胞(CT26.WT)、成纤维细胞(L929)、胃癌细胞(MFC)及腹水癌细胞(S180),发现均有极显著的杀伤效果(P<0.01),且对肺癌细胞(A549)的杀伤效果最好,当HM-P 终浓度为20 μg/mL 时,杀伤率高达42.6%。
Abstract:To determine the structure composition and in vitro anti-tumor activity of Hypsizygus marmoreus polysaccharide (HM-P), HM-P was isolated and extracted by hot water extraction method. The HM-P components were identified by gel permeation chromatography, Fourier transform infrared spectroscopy, and high performance liquid chromatography. The in vitro anti-tumor activity of HM-P was examined by cell counting kit-8(CCK-8) and cell scratch method. The result showed that HM-P, with a weight average molecular weight(MW) of 25 232 Da, was composed of galactose, glucose, and xylose in the content ratio of 72.406∶17.661∶9.933. HM-P could significantly inhibit the proliferation (P<0.05) and migration (P<0.05) of lung cancer cells (A549), colon cancer cells (CT26.WT), fibroblasts (L929), and mouse forestomach carcinoma(MFC).It could also highly significantly inhibit the proliferation of ascites cancer cells (S180) (P<0.01). The direct inhibition of HM-P on mouse forestomach carcinoma(MFC) was the strongest. When the final concentration of HM-P was 20 μg/mL, the inhibition rate reached 27.9% and the mobility rate 19.0%. HM-P mediated the supernatant of macrophages (RAW264.7) to kill lung cancer cells (A549), colon cancer cells (CT26.WT), fibroblasts (L929), mouse forestomach carcinoma (MFC) and ascites cancer cells (S180). It was found that the killing effect was extremely significant (P<0.01), especially on lung cancer cells (A549). The killing rate of lung cancer cells (A549) was 42.6% when the final concentration of HM-P was 20 μg/mL.
