干巴菌不同溶剂提取物功能成分与生物活性

中文摘要: 为进一步挖掘干巴菌的生物活性成分,采用6 种不同溶剂(水、乙醇、丙酮、乙酸乙酯、氯仿、石油醚)提取干巴菌,比较不同溶剂提取物的总酚和麦角甾醇含量,分析每种提取物所含的多酚组分,并探究提取物的抗氧化性及其对α-葡萄糖苷酶的抑制活性。结果表明:6 种提取物中乙醇提取物的总酚含量最高,为(14.95±0.58) mg/g,不同溶剂提取物所含的多酚组成及含量差异明显。乙酸乙酯和乙醇提取物中麦角甾醇含量高于其他溶剂提取物,分别为(1.33±0.13) mg/g 和(1.15±0.15) mg/g。干巴菌乙醇提取物的抗氧化性较好,其清除DPPH 自由基和ABTS+自由基的能力最强,铁还原能力最强。此外,丙酮、乙醇和乙酸乙酯提取物均对α-葡萄糖苷酶表现出明显的抑制能力。相关性分析发现干巴菌所含的酚类化合物、麦角甾醇等成分是其具有抗氧化性及α-葡萄糖苷酶抑制活性的潜在物质。
Abstract:To further explore the bioactive ingredients of Thelephora ganbajun, six different solvents were used to extract Thelephora ganbajun, including water, ethanol, acetone, ethyl acetate, chloroform and petroleum ether. The effects of different solvents on the content of total phenols and ergosterol were investigated. At the same time, the polyphenol fractions, the antioxidant activity of the extracts and its inhibitory effect on the activity of α-glucosidase were analyzed. The result indicated that among the six extracts, the ethanol extract had the highest content of total phenols, which was (14.95±0.58) mg/g. The polyphenol components and content among different solvent extracts obviously varied. The content of ergosterol in ethyl acetate and ethanol extracts, (1.33±0.13) and (1.15±0.15) mg/g, respectively, were higher than that in other solvent extracts. The ethanol extract had the best antioxidant activity with the strongest scavenging ability of DPPH and ABTS+ free radicals, as well as the strongest ferric ion reducing antioxidant power. In addition, acetone, ethanol and ethyl acetate extracts showed significant α-glucosidase inhibitory activity. According to correlation analysis, Thelephora ganbajun contained polyphenol components and ergosterol, potential substances for its antioxidant activity and α-glucosidase inhibitory activity.
