中文摘要: 为研究主要存在于烟草、罗勒精油和薰衣草精油中的芳香化合物——罗勒烯,在减轻中波紫外线(ultraviolet B,UVB)诱导的HaCaT 角质形成细胞和Hs68 成纤维细胞光老化中的潜在益处。通过检测氧化应激损伤标志物和基质金属蛋白酶(matrix metalloproteinases,MMP)的表达水平,评估罗勒烯缓解表皮HaCaT 细胞光老化的作用。通过测定MMP 的分泌和细胞外基质的组成,评估罗勒烯缓解真皮Hs68 细胞光老化的效果。结果显示,在暴露于UVB 的表皮HaCaT 细胞中,罗勒烯能有效减少活性氧和丙二醛的积累,提高超氧化物歧化酶的活性,从而缓解光老化引起的氧化应激。此外,罗勒烯还能抑制UVB 诱导的MMP 表达水平升高。在暴露于UVB 的真皮Hs68 细胞中,罗勒烯通过抑制MMP 的表达和促进胶原蛋白及透明质酸的分泌,发挥保护细胞外基质的作用,表明罗勒烯对皮肤光老化存在保护作用。
Abstract:This study investigated the potential benefits of ocimene, an aromatic compound predominantly present in tobacco, basil essential oil, and lavender essential oil, in mitigating the effect of ultraviolet B(UVB)-induced photoaging in HaCaT keratinocytes and Hs68 fibroblasts. The photoaging-mitigating effect of ocimene in epidermal HaCaT cells was assessed by measuring oxidative stress damage markers and matrix metalloproteinase (MMP) expression levels. The photoaging-mitigating effect of ocimene in dermal Hs68 cells was evaluated by measuring the secretion of MMP and the composition of the extracellular matrix. The results showed that in UVB-exposed epidermal HaCaT cells, ocimene effectively reduced the accumulation of reactive oxygen species and malondialdehyde but increased the activity of superoxide dismutase. The photoaging-induced oxidative stress was thus alleviated. Moreover, ocimene restrained the rise of UVB-induced MMP expression levels. In UVB-exposed dermal Hs68 cells, ocimene exerted a protective effect on the extracellular matrix by inhibiting MMP expression and promoting collagen and hyaluronic acid secretion, indicating its protective effect on skin photoaging.
