白及多糖单糖组成及其抑制酪氨酸酶动力学机制

中文摘要: 该文采用硫酸-苯酚法测定白及多糖含量,通过离子色谱法测定白及多糖单糖组成,使用酶标仪得到白及多糖抑制酪氨酸酶活力系列动力学参数。结果表明,复合酶法提取白及多糖含量为(51.97±1.52)%。离子色谱法得到白及多糖单糖组成为鼠李糖、阿拉伯糖、半乳糖、葡萄糖、甘露糖、半乳糖醛酸和葡萄糖醛酸共7 种,其单糖组成比例分别为0.17%、0.36%、1.86%、41.81%、54.21%、0.71%和0.88%。白及多糖对酪氨酸酶抑制作用的IC50为59.31 mg/mL,白及多糖可以降低酪氨酸酶反应初速度,表明白及多糖对酪氨酸酶的催化作用存在迟滞影响,且随着白及多糖质量浓度的增加,对酪氨酸酶活力的抑制作用越强,具有剂量依赖性。此外,白及多糖对酪氨酸酶的抑制作用具有可逆性,Lineweaver-Burk 双倒数曲线图结果显示,随着白及多糖质量浓度的增加,米氏常数Km 值整体逐渐增大,而Vmax 值整体逐渐降低,表明白及多糖对酪氨酸酶的抑制作用表现为非竞争性与竞争性混合的抑制类型。
Abstract:The content of Bletillɑ striɑtɑ polysaccharides was determined by the phenol-sulfuric acid method,the monosaccharide composition was measured by the ion chromatography method, and the dynamic parameters of inhibiting tyrosinase activity of Bletillɑ striɑtɑ polysaccharides were obtained by adopting the enzymolysis assay. The results showed that the content of Bletillɑ striɑtɑ polysaccharides extracted by the multi-enzyme method was (51.97±1.52)%. By employing ion chromatography, the study found that the monosaccharide composition of Bletillɑ striɑtɑ polysaccharides included seven kinds: Rha, Ara, Gal, Glc, Man, GAL-UA and GLc-UA, and the proportions of monosaccharides were 0.17%, 0.36%, 1.86%, 41.81%, 54.21%, 0.71%,and 0.88% respectively. In vitro, Bletillɑ striɑtɑ polysaccharides exerted an inhibitory effect on tyrosinase’s activity, with the IC50 of Bletillɑ striɑtɑ polysaccharides being 59.31 mg/mL. Additionally, Bletillɑ striɑtɑ polysaccharides could reduce the initial reaction speed of tyrosinase, which indicated that Bletillɑ striɑtɑ polysaccharides had a delayed effect on the catalysis of tyrosinase. Meanwhile, with the increasing mass concentration of Bletillɑ striɑtɑ polysaccharides, they had a stronger inhibition effect on tyrosinase′s activity, with the characteristics of dose-dependency and reversibility. The Lineweaver-Burk double-reciprocal curve showed that with the rising mass concentration of Bletillɑ striɑtɑ polysaccharides, the value of the Michaelis constant Km gradually increased, while Vmax gradually decreased. This indicated that the inhibitory effect of Bletillɑ striɑtɑ polysaccharides on tyrosinase was a type of non-competitive and competitive mixed inhibition.
