中文摘要: 以滇橄榄为原料,基于单因素试验结果,采用响应面曲面法对滇橄榄多酚氧化酶(polyphenol oxidase,PPO)提取工艺进行优化,结果表明:回归模型对响应值拟合良好,得出滇橄榄PPO 提取最佳工艺条件为磷酸盐缓冲液pH7.9、浸提时间1.5 h、液料比4.0 ∶1(mL/g),在该条件下,模型预测滇橄榄PPO 酶活力为24.83 U/(g·min)。并在该基础上对滇橄榄PPO 抑制剂进行筛选,得出柠檬酸亚锡二钠的抑制作用最强,且浓度为0.6 g/L 的柠檬酸亚锡二钠在对滇橄榄PPO 作用时间160 min 及以上时,PPO 活性能够被有效抑制。

Abstract:Based on the results of univariate analysis,the extraction process of polyphenol oxidase(PPO)from Phyllanthus emblica L.was optimized by the response surface method.The regression model fit the response values well.The optimal process conditions for the extraction of P.emblica L.PPO were phosphate buffer pH of 7.9,extraction time of 1.5 h,and liquid-to-material ratio of 4.0 ∶1(mL/g).Under these conditions,the model predicted the activity of P.emblica L PPO to be 24.83 U/(g·min).In addition,inhibitors of P.emblica L.PPO were screened.Disodium stannous citrate had the strongest inhibitory effect;this effectively inhibited the PPO activity at a concentration of 0.6 g/L,when the action time of PPO was 160 min or more.
