黄药叶总黄酮提取工艺优化及其抗氧化活性研究

中文摘要: 采用超声波辅助提取法提取黄药叶总黄酮,通过单因素试验和正交试验优化黄药叶总黄酮提取工艺,并探讨其体外抗氧化活性。结果表明,黄药叶总黄酮最佳提取工艺参数为乙醇体积分数70%,提取次数3次,提取时间40 min,料液比1∶30(g/mL),此条件下黄药叶总黄酮得率为(6.18±0.13)%。体外抗氧化试验结果表明,黄药叶总黄酮提取物对羟自由基、DPPH自由基和超氧阴离子自由基均具有一定的清除能力,相应的半数清除浓度(inhibitory concentration,IC50)为 484.12、18.27、46.88 μg/mL,分别为 L-抗坏血酸的 1.50、4.50、2.51 倍,黄药叶总黄酮提取物铁离子还原能力略低于L-抗坏血酸。
Abstract:Single factor test and orthogonal test were used to optimize ultrasound-assisted extraction of total flavonoids from Premna cavaleriei Levl.leaves.The antioxidant activity in vitro of the leaves was analyzed.Optimized extraction parameters were established as follows:ethanol concentration 70%;extraction times 3;extraction time 40min;solid/liquidratio 1∶30(g/mL).Under these conditions,total flavonoid yield was(6.18 ±0.13)%.The results of antioxidant experiments showed that the total flavonoids extracted from Premna cavaleriei Levl.leaves had certain scavenging rate of hydroxyl radicals,DPPH radicals and superoxide anion radicals.The corresponding inhibitory concentration(IC50)was 484.12,18.27 and 46.88 μg/mL,respectively and were 1.50,4.50 and 2.51 times that of L-ascorbic acid,respectively.The ferric ion reducing ability of total flavonoids from Premna cavaleriei Levl.leaves was slightly lower than that of L-ascorbic acid.
