中文摘要: 通过高效液相色谱(high performance liquid chromatography,HPLC)法测定藏红花中有效成分西红花苷-Ⅰ和西红花苷-Ⅱ的含量;通过紫外-可见分光光度法测定标示藏红花香味强度、芳香度和色度的3 个特征参数,以评价国内外不同来源藏红花的品质。采用十八烷基硅烷键合硅胶色谱柱,流动相为甲醇-水(45∶55,体积比),流速1 mL/min,检测波长 440 nm,柱温 30 ℃。西红花苷-Ⅰ、西红花苷-Ⅱ分别在 5.419 μg/mL~108.373 μg/mL(r=1.000 0)和 3.822 μg/mL~76.433 μg/mL(r=0.999 9)范围内线性关系良好,平均加样回收率分别为99.81%(相对标准偏差为1.50%)和97.47%(相对标准偏差为0.95%)。运用该HPLC 法测定,结果准确、可靠,国内外不同来源藏红花中有效成分含量、品质差异显著。紫外-可见分光光度法测定波长分别为257、330、440 nm,不同来源藏红花的3 个特征参数具有显著差异,分属不同质量等级。
Abstract:In order to evaluate the quality of saffron from different sources at home and abroad,a high performance liquid chromatography(HPLC)method was established to determine the content of crocinⅠ and crocinⅡ,meanwhile,a UV/visible spectrophotometry was established to determine the three characteristic parameters of odor,flavor and color in saffron.HPLC method was carried out with column C18,using methanol and water(45∶55,volume ratio)as the mobile phase at flow rate of 1 mL/min.The detection wavelength was at 440 nm and the column temperature was at 30 ℃.The calibration curve of crocinⅠ was in good linearity at 5.419 μg/mL-108.373 μg/mL (r=1.000 0)and the average recovery was 99.81 % with relative standard deviation(RSD)of 1.50%.The calibration curve of crocinⅡ was in good linearity at 3.822 μg/mL-76.433 μg/mL(r=0.999 9)and the average recovery was 97.47 % with RSD of 0.95 %.The HPLC method was accurate and stable.There was a significant difference in the content of crocinⅠ and crocinⅡ.UV/visible spectrophotometry was carried out at 257,330 nm and 440 nm.The absorbance values of three characteristic parameters in saffron were different.The saffron samples belonged to the different grade.
