中文摘要: 该文对产L-谷氨酸的谷氨酸棒杆菌(Corynebacterium glutamicum)过表达海藻糖合酶进行研究。利用组成型表达质粒在产L-谷氨酸 C. glutamicum CICC20935 中过表达海藻糖合酶获得重组菌株20935T。结果显示,菌株20935T 在以葡萄糖为碳源的培养过程中呈现出絮凝表型并且L-谷氨酸产量提高70%。超薄切片透射电子显微镜分析结果显示,菌株20935T 细胞壁外侧含有海藻糖的分枝菌酸酯膜层完整性降低。在菌株20935T 培养过程中添加麦芽糖可抑制海藻糖合酶将海藻糖转化为麦芽糖,菌体不会出现絮凝表型,同时L-谷氨酸产量降低35%~48%。过表达海藻糖合酶能够影响C. glutamicum 细胞壁完整性和L-谷氨酸产量。
Abstract:The overexpression of trehalose synthase in the L-glutamate producer Corynebacterium glutamicum was studied. The constitutive expression plasmid was used for overexpression of trehalose synthase in Lglutamate producing C. glutamicum CICC20935 and the recombinant strain 20935T was obtained. The strain 20935T displayed a flocculation phenotype and increased the L-glutamate production by 70% in the medium with glucose as the sole carbon source. The results of ultrathin section transmission electron microscopy revealed that the integrality of trehalose-containing outer membrane mycomembrane of the cell wall of strain 20935T was decreased. Addition of maltose in the culture process of strain 20935T suppressed the conversion of trehalose to maltose by trehalose synthase,and the cells did not display the flocculation phenotype,with the L-glutamate production decreasing by 34%-48%. Overexpression of trehalose synthase in C. glutamicum could affect cell wall integrity and L-glutamate production.
