中文摘要: 该文从青岛海泥中筛选产普鲁兰酶菌株,通过形态学、生理生化特征、16S rRNA 进行分析鉴定,研究酶学性质并用该酶处理12 个不同来源(荞麦、玉米、红薯、小麦、大麦、粘米、水磨糯米、百合、蚕豆、葛根、绿豆、山药)淀粉。筛选得到产Ⅱ型普鲁兰酶的菌株Bacillus sp. A1,该酶的最适反应温度为40 ℃,最适pH 值为5,在反应温度45 ℃和55 ℃下保温2 h,相对酶活可达70%以上,在pH5~7 缓冲液中稳定性高,表明该酶具有较好的热稳定性和pH 值稳定性。经酶处理的荞麦、玉米和红薯淀粉可形成均匀、深入内部的孔隙。多孔淀粉的溶解度和膨胀力相较于天然淀粉平均增加约2.11% 和2.25%;酶解24 h 的多孔淀粉的吸水率和吸油率相较于天然淀粉平均增加约44.80% 和30.11%。综上,该酶在荞麦、玉米和红薯多孔淀粉的制备中具有一定的应用前景。
Abstract:This study aimed to isolate pullulanase-producing strains from Qingdao marine mud. Through morphological,physiological,biochemical,and 16S rRNA analyses,the strain's enzymatic properties were investigated. The enzyme was then used to treat starches from 12 different sources(buckwheat,corn,sweet potato,wheat,barley,glutinous rice,water-milled glutinous rice,lily,broad bean,kudzu root,mung bean,and yam). A strain producing typeⅡpullulanase,Bacillus sp. A1,was obtained. The optimal reaction temperature and pH for the enzyme were 40 ℃ and 5,respectively. After incubation at 45 ℃ and 55 ℃ for 2 h,the relative enzyme activity remained above 70%. The enzyme exhibited high stability in buffers with pH values ranging from 5 to 7,indicating good thermal and pH stability. Treatment with the enzyme resulted in the formation of uniform and deeply penetrating pores in buckwheat,corn,and sweet potato starches. Compared with those of native starches,the solubility and swelling power of the porous starches increased by approximately 2.11% and 2.25%,respectively. After 24 h of enzymatic hydrolysis,the water and oil absorption capacities of the porous starches increased by approximately 44.80% and 30.11%,respectively,compared with those of native starches.In summary,this enzyme shows potential for application in the preparation of porous starches from buckwheat,corn,and sweet potato.
