植物乳植杆菌LZ026 降脂作用及其对小鼠肠道菌群的影响

中文摘要: 为从贵州农家酸肉中筛选并鉴定乳酸菌,研究其降脂作用及对肠道菌的调节作用。通过稀释涂布平板法及菌株16S rDNA 序列分析鉴定出一株植物乳植杆菌LZ026。通过高脂饮食诱导小鼠,结合阴性对照和辛伐他汀(simvastatin)阳性对照,分析植物乳植杆菌LZ026 对高脂小鼠脂质代谢和肠道菌群的影响。实验终点后,使用单试剂甘油磷酸氧化酶-过氧化物酶偶联法和双试剂直接法检测小鼠血清和肝脏中脂质相关指标含量,采用苏木精-伊红染色结肠组织观察其形态变化,采用16S rDNA 高通量测序分析肠道菌群组成变化。结果表明,植物乳植杆菌LZ026 可以显著降低高脂小鼠的体质量(p<0.05),减少肝脏指数、肾脏指数和脾脏指数,降低小鼠血清和肝脏中的总胆固醇、甘油三酯和低密度脂蛋白含量,增加高密度脂蛋白含量,增加小鼠结肠绒毛长度和隐窝深度。门水平上增加拟杆菌门(Bacteroidetes)和放线菌门(Actinobacteria)相对丰度,降低厚壁菌门(Firmicutes)相对丰度;科水平上增加乳酸杆菌科(Lactobacilaceae)和普雷沃氏菌科(Prevotellaceae)相对丰度;属水平上,灌胃植物乳植杆菌后乳杆菌属(Lactobacillus)相对丰度相应增多。由此表明,植物乳植杆菌LZ026 具有降低脂肪水平和调节肠道菌群的作用。
Abstract:Lactic acid bacteria were screened and identified from Guizhou farmhouse sour meat and their lipidlowering effects and regulatory effects on intestinal microbiota in mice were investigated. A strain of Lactiplantibacillus plantarum LZ026 was identified through the dilution coating plate method and 16S rDNA sequence analysis. Mice were induced with a high-fat diet, and a negative control group and a positive one treated with simvastatin were included. The effects of Lactiplantibacillus plantarum LZ026 on lipid metabolism and intestinal flora in high-fat mice were analyzed. After the experimental endpoint, the content of lipid-related indicators in the serum and liver of the mice was detected by the single-reagent glycerol-3-phosphate oxidase-peroxidase method and the double-reagent direct method. The morphological changes of colon tissue were observed by hematoxylineosin staining staining, and the changes in the composition of intestinal flora were analyzed by 16S rDNA high-throughput sequencing. According to the results, Lactiplantibacillus plantarum LZ026 could significantly reduce the body weight of high-fat mice (p<0.05), lower the indicators of liver, kidney, and spleen, decrease the content of total cholesterol, triglyceride, and low-density lipoprotein while adding highdensity lipoprotein content in both serum and liver of high-fat mice. Colon villus length and crypt depth were also increased. At the phylum level, relative abundances of Bacteroidetes and Actinobacteria rose while Firmicutes declined. At the family level, relative abundances of Lactobacilaceae and Prevotellaceae increased. At the genus level, Lactobacillus abundance went higher after the gastric gavage of Lactiplantibacillus plantarum.These findings demonstrate that Lactiplantibacillus plantarum LZ026 exhibits a lipid-lowering effect and regulates intestinal microbiota.
