中文摘要: 该研究探究内蒙古呼和浩特和通辽地区蒙古族人肠道中双歧杆菌(Bifidobacterium)的数量和生物多样性特点,并探讨地域对人体肠道内双歧杆菌数量和生物多样性的影响。以内蒙古呼和浩特地区和通辽地区各50 名蒙古族成年志愿者为研究对象,借助双歧杆菌特异性引物,采用PacBio Sequel II 测序技术和微滴式数字聚合酶链式反应(droplet digital polymerase chain reaction,dd-PCR)技术解析志愿者肠道中双歧杆菌的数量和生物多样性特点。结果表明,呼和浩特地区志愿者肠道内双歧杆菌的平均生物量[(1.64×108±2.16)CFU/g]小于通辽地区[(2.04×108±3.72 )CFU/g];在内蒙古志愿者肠道内共注释到11 种双歧杆菌,两地区志愿者肠道中双歧杆菌多样性和结构无显著差异,但角双歧杆菌(B.angulatum)和长双歧杆菌(B.longum)等在呼和浩特市志愿者肠道中富集(P<0.05),齿双歧杆菌(B.dentium)、链状双歧杆菌(B.catenulatum)以及热嗜酸性双歧杆菌(B.thermacidophilum)等在通辽市志愿者中显著增加。Spearman 分析结果发现,呼和浩特市志愿者肠道内双歧杆菌间互作网络更密集(P<0.05,|R|>0.3)。相比于多样性和结构,地域因素显著影响了肠道双歧杆菌的相对丰度和互作网络关系。
Abstract:This study investigated the quantity and biodiversity of Bifidobacterium in the intestines of Mongolian people in Hohhot and Tongliao, Inner Mongolia, and explored the influence of region on the quantity and biodiversity of Bifidobacterium in people's intestines. With 50 Mongolian adult volunteers from Hohhot and Tongliao, Inner Mongolia as the research objects, specific primers of Bifidobacterium were used. The number and biodiversity of Bifidobacterium in the intestines of volunteers were analyzed by PacBio Sequel II sequencing technology and droplet digital polymerase chain reaction (ddPCR) technology. The average biomass of Bifidobacterium in the intestine of volunteers from Hohhot[(1.64×108±2.16)CFU/g]was lower than that of volunteers from Tongliao [(2.04×108±3.72 )CFU/g]. A total of 11 species of Bifidobacterium were annotated in the intestine of volunteers from Inner Mongolia. There was no significant difference in the diversity and structure of Bifidobacterium in the intestine of volunteers from the two regions. However, B. angulatum, and B. longum were enriched in the intestines of volunteers from Hohhot (P<0.05), while B. dentium, B. catenulatum, B. biavatii, and B. thermacidophilum were significantly increasing in those of volunteers from Tongliao. According to Spearman analysis, the interaction network between Bifidobacterium in the intestines of volunteers from Hohhot was denser (P<0.05, |R|>0.3). Compared with diversity and structure, geographical factors significantly affect the relative abundance and interaction network relationship of intestinal Bifidobacterium.
