中文摘要: 该研究从海泥样品中筛选产普鲁兰酶的微生物。采用透明圈法从青岛海泥中筛选到产普鲁兰酶的菌株A2,通过菌株形态特征和16S rDNA 分析,鉴定菌株A2 为芽孢杆菌属(Bacillus sp.),其生长的最适碳源、氮源、生长温度和pH 值分别为酵母浸膏、大豆蛋白胨、40 ℃和7.0;产酶的最适碳源、氮源、温度和初始pH 值分别为蔗糖、大豆蛋白胨、45 ℃和7.0。普鲁兰酶的最适温度和初始pH 值分别为35 ℃和6.0,在35~50 ℃、pH5.0~9.0 范围具有良好的稳定性;Ba2+、NH4+、Zn2+、K+、Mg2+、Cu2+、Ca2+和Na+等金属离子对酶活影响小。用该酶水解不同种类淀粉,可观察到玉米淀粉表面形成均匀的多孔形态,该酶可用于制备玉米多孔淀粉。
Abstract:The microorganisms producing pullulanase were screened from marine samples. A strain A2 producing pullulanase was isolated from the sea mud collected from Qingdao by the transparent circle method. Strain A2 was identified as Bacillus sp. based on the morphological characteristics and 16S rDNA sequence. The optimum carbon source, nitrogen source, temperature, and pH value for the growth of the strain were yeast extract, soybean peptone, 40 ℃, and 7.0, respectively, and those for enzyme production of the strain were sucrose, soybean peptone, 45 ℃, and 7.0, respectively. The optimum temperature and pH value of pullulanase were 35 ℃ and 6.0, respectively, and the pullulanase demonstrated good stability within the ranges of 35-50 ℃ and pH5.0-9.0. Metal ions such as Ba2+, NH4+, Zn2+, K+, Mg2+, Cu2+, Ca2+, and Na+ had little effects on the enzyme activity. When different kinds of starch were hydrolyzed by this enzyme, uniform porous morphology was observed on the surface of corn starch. The enzyme can be used to prepare porous starch of maize.
