中文摘要: 该文以特殊医学用途配方奶粉为原料,旨在优化和确定超高效液相色谱-串联质谱法检测该产品中5 种核苷酸的最佳方法。通过优化前处理方法和仪器条件,最终前处理方法为取样量0.80 g,溶剂温度40 ℃,用量40 mL,酸碱调节剂为25%的乙酸溶液,调节最终pH 值为4.2±0.1;优化液相色谱、质谱条件和最佳检测条件:电喷雾电源离子模式,多反应监测扫描方式,5 mmol/L 乙酸铵-0.1%甲酸与乙腈作为流动相进行梯度洗脱,在该条件下5 种核苷酸的平均回收率为91.865%~100.815%。该方法能够快速准确地测定特殊医学用途配方奶粉中5 种核苷酸含量。
Abstract:This article used special medical formula milk powder as the raw material to optimize and determine the best method for detecting five nucleotides in this product by using ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).By optimizing the pre-processing methods and instrument conditions,the final pre-processing method was adopted under the following conditions:0.80 g of sample was taken,and the solvent temperature was 40 ℃.40 mL of solvent was used,with a pH regulator of 25% acetic acid solution to adjust the final pH to 4.2±0.1.The UPLC-MS/MS and detection conditions were optimized as follows:electrospray ionization and ion mode and multiple response monitoring scans were used,and 5 mmol/L ammonium acetate-0.1% formic acid solution and acetonitrile was used as mobile phase for gradient elution.Under these conditions,the average recovery rates for the five nucleotides ranged from 91.865% to 100.815%.This method could rapidly and accurately determine the content of the five nucleotides in special medical formula milk powder.
