中文摘要: 采用单因素试验以及响应面法优化复合酶法提取诺丽多糖的工艺,探讨提取时间、料液比、提取温度、pH 值对多糖提取率的影响。通过紫外光谱、红外光谱扫描技术对诺丽多糖的结构进行分析,并检测其体外抗氧化作用。结果表明:诺丽多糖复合酶法最佳提取工艺为提取时间1.9 h、料液比1∶39( g/mL)、提取温度62 ℃、pH5.5。经光谱扫描鉴定,该工艺下的诺丽多糖含有少量蛋白质,具有典型的多糖结构,是一种酸性多糖。体外抗氧化试验证明,诺丽多糖对DPPH 自由基、ABTS+自由基、羟自由基、超氧阴离子自由基均有较强的清除能力,在抗氧化方面有良好的效果。
Abstract:In this study, complex enzyme extraction of noni (Morinda citrifolia Linn.) polysaccharides was optimized. The influences of extraction time, solid-liquid ratio, extraction temperature, and pH on polysaccharide extraction rate were discussed. The extraction process of noni polysaccharide was optimized by a single factor test and response surface method. The structure of noni polysaccharide was analyzed by UV and infrared scanning techniques and its antioxidant effect in vitro was also analyzed. The results showed that the best extraction process was 1.9 h, solid-liquid ratio 1∶39 (g/mL), extraction temperature 62 ℃, and pH5.5. It was identified from spectral scanning identification that the noni polysaccharide produced in this process containing a small amount of protein in a typical polysaccharide structure was an acidic polysaccharide. The in vitro antioxidant test proved that the scavenging capacity of noni polysaccharides was relatively strong on radicals including DPPH·, ABTS+·,·OH, and O2-·. Therefore, noni polysaccharides showed good antioxidant effect.
