中文摘要: 利用单因素试验、正交试验结合验证试验,以乙醇浓度(50%、60%、70%、80%、90%)、料液比[1∶15、1∶20、1∶25 、1∶30、1∶35( g/mL)]、提取时间(20、40、60、80、100 min)筛选莓茶活性物质的最佳提取条件,研究莓茶提取物对哈维氏弧菌抑菌活性影响;采用牛津杯法测定最小抑菌浓度;通过生长曲线、细胞膜完整性和通透性、细胞壁以及扫描电镜观察等方法探讨莓茶提取物的抑菌作用及机理。结果表明:莓茶提取物最佳提取条件为乙醇浓度80%、料液比1∶30( g/mL)、提取时间60 min。基于此条件莓茶提取物对哈维氏弧菌的抑菌圈直径为(23.67±0.17) mm,莓茶提取物的最小抑菌浓度为0.015 6 g/mL。经最小抑菌浓度(minimal inhibit concentration, MIC)处理后,发现莓茶提取物对哈维氏弧菌生长曲线抑制效果好;能够破坏菌体细胞膜完整性,增大细胞膜的通透性,使细胞内生物大分子渗出;对哈维氏弧菌细胞壁有明显破坏作用;菌体表面变粗糙,出现裂解胞浆外流,导致细胞边缘模糊,这表明莓茶提取物对哈维氏弧菌具有较强抑制作用。
Abstract:Using the single factor test, the orthogonal test in combination with the verification test, the optimum extraction process of flavonoids (dihydromyricetin, myricetin, myricetin) from Ampelopsis grossedentata was studied in terms of the ethanol solvent concentration (50%, 60%, 70%, 80%, 90%), the solid/liquid ratio [1∶15, 1∶20, 1∶25, 1∶30, 1∶35 (g/mL)], extraction time (20, 40, 60, 80, 100 min). The effect of A.grossedentata extract on the antibacterial activity of Vibrio harveyi was thus investigated. The minimum inhibitory concentration was determined with the Oxford cup method. Growth curve, cell membrane integrity and permeability, cell wall, and scanning electron microscope were applied to discuss the antibacterial activity and mechanism of A. grossedentata extract. The result showed that the optimum extraction conditions were an ethanol concentration of 80%, a solid/liquid ratio of 1∶30(g/mL), and an extraction time of 60 minutes. Under these conditions, the diameter of the inhibitory zone of the A. grossedentata extract against V. harveyi was(23.67±0.17) mm and the minimum inhibit concentration of the A. grossedentata extract was 0.015 6 g/mL. After minimum inhibitory concentration(MIC) treatment, the A. grossedentata extract was found to have a good inhibitory effect on the growth curve of V. harveyi. It could destroy the integrity of the cell membrane, increase the permeability of the cell membrane, and cause the release of intracellular biomacromolecules. The cell wall of V. harveyi was damaged. The surface of the bacteria became rough, the lysed cytoplasm flowed out and the edge of the cells was blurred, indicating that the A. grossedentata extract had a strong inhibitory mechanism against V. harveyi.
