超声辅助提取长根菇多酚及其抗氧化活性

中文摘要: 多酚是一类具有生物活性的天然化合物。为进一步发掘长根菇多酚的应用价值,该研究以长根菇为原料,采用超声辅助提取多酚,采用响应面试验对其提取工艺参数进行优化,并考察长根菇多酚的体外抗氧化活性。研究结果表明,长根菇多酚的最佳提取工艺为乙醇浓度45%、超声时间65 min、液料比50∶1(mL/g),在该条件下,长根菇多酚的得率达到9.82 mg/g;体外抗氧化研究表明,长根菇多酚具有较好的2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS]阳离子自由基、·OH 和 1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基的清除能力,且呈现剂量依赖性,其IC50分别为 280、110.78 μg/mL和178.78 μg/mL。

Abstract:Polyphenols are a group of natural compounds with biological activity.This study aimed to further explore the application value of polyphenols from Oudemansiella radicata.To be specific,ultrasonic-assisted extraction of polyphenols from O.radicata was carried out,and the in vitro antioxidant activity of the polyphenols was investigated.The results showed that the optimal extraction process was 45%ethanol,sonication for 65 min,and liquid-to-material ratio of 50 ∶1(mL/g).With this extraction process,the yield of polyphenols was up to 9.82 mg/g,and the polyphenols can well scavenge 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)cation free radical,·OH,and 1,1-diphenyl-2-picrylhydrazyl(DPPH)in a dose-dependent manner,with half-maximal inhibitory concentration(IC50)of 280,110.78 μg/mL,and 178.78 μg/mL,respectively.
