中文摘要: 比较研究西藏新鲜曲拉和干燥曲拉细菌多样性差异。通过Illumina MiSeq测序技术,分析西藏林芝产新鲜牦牛曲拉和干燥牦牛曲拉细菌16SrDNA V1-V3区基因序列,对不同状态牦牛曲拉细菌群落结构进行系统分析和比较。结果表明新鲜曲拉(XZL)获得(29 566.67±1 703.40)条高质量序列,聚成(594.33±36.26)个可操作分类单元(operational taxonomic units,OTUs),干燥曲拉(GLZ)获得(33 604.67±2 042.39)条高质量序列,聚成(690.67±56.82)个 OTUs。新鲜曲拉和干燥曲拉在门水平上主要细菌群落隶属于厚壁菌门(Firmicutes)和变形菌门(Proteobacteria),在属水平上中新鲜曲拉检测出72个属,干燥曲拉检测出96个属,二者主要优势乳球菌属(Lactococcus)、沙雷氏菌属(Serratia)、不动杆菌属(Acinetobacter)。干燥曲拉的Alpha多样性指数相关指标辛普森指数(Simpson)指数的平均值为0.90±0.01、Chao1指数平均值为760.21±133.80、ACE指数平均值为 775.91±116.98、香浓指数(Shannon)指数平均值为 5.46±0.05,而新鲜曲拉的Simpson指数平均值为0.90±0.01、Chao1指数平均值为595.31±45.23、ACE指数平均值为602.15±49.33、Shannon指数平均值为5.39±0.17,由此可知与新鲜曲拉相比干燥曲拉细菌多样性更为丰富。
Abstract:A comparative study was conducted on the differences of bacterial diversity in the fresh Qula(XZL)and dried Qula(GLZ)in Tibet.In this study,the 16S rDNA V1-V3 regions of fresh Qula sample and dried Qula sample from Nyingchi were sequenced by high-throughput sequencing technology to analyse the diversity and community structure of bacteria systematicly.The results showed that 29566.67±1703.40 high-quality sequences and 594.33±36.26 OTUs were obtained from XZL,33604.67±2042.39 high-quality sequences and 690.67±56.82 OTUs were obtained from GLZ.Firmicutes and Proteobacteria were the dominant phylum in different samples at the phylum level.72 genus and 96 genus were obtained in XZL and GLZ,and the dominant bacterial genera in the two samples were Lactococcus,Serratia and Acinetobacter at the genus level.The Simpson index,Chao1 index,ACE index,and Shannon index were used to evaluate the alpha diversity of the samples,and the results showed that the Simpson index,Chao1 index,ACE index,and Shannon index were 0.90±0.01,760.21±133.80,775.91 ±116.98,5.46 ±0.05 and 0.90±0.01,595.31±45.23,602.15 ± 49.33,5.39±0.17 in GLZ and XZL respectively,and the results indicated that the GLZ had a relatively high abundance.
