中文摘要: 为探究副干酪乳杆菌(Lacticaseibacillus paracasei)SMR-0128的发酵工艺及其对肠道动力的作用,通过单因素试验和响应面试验设计优化副干酪乳杆菌SMR-0128的发酵培养条件,利用动物便秘模型,分析其对小鼠润肠通便功能的影响。确定最佳发酵条件为装液量140 mL/250 mL、接种量4%、菌株种龄12 h、转速74 r/min、培养基初始pH值为7、36 ℃培养25 h。在此条件下菌体量达到3.22×1010 CFU/mL,相比优化培养条件前提高12.5倍。实验动物每日服用菌体量为1×109、1×108 CFU/kg bw的副干酪乳杆菌SMR-0128,与对照组相比,实验组小鼠的小肠推进率明显提高、首粒排黑便时间明显缩短,表明副干酪乳杆菌SMR-0128具有良好的润肠通便功能。
Abstract:To investigate the fermentation process of Lacticaseibacillus paracasei SMR-0128 and its effects on intestinal motility,the fermentation culture conditions of Lacticaseibacillus paracasei SMR-0128 were optimized by single-factor experiments and response surface methodology (RSM),and its effects on the intestinemoistening and laxative functions of mice were analyzed using an animal constipation model. The optimal fermentation conditions were determined as follows:liquid volume of 140 mL/250 mL,inoculum amount of 4%,seed age of 12 h,rotation speed of 74 r/min,initial pH value of 7,and culture at 36 ℃ for 25 h. Under these conditions,the biomass reached 3.22×1010 CFU/mL,which was 12.5 times higher than that before optimization.The experimental animals were administered Lacticaseibacillus paracasei SMR-0128 at daily doses of 1× 109 CFU/kg bw and 1×108 CFU/kg bw. Compared with the control group,the small intestinal propulsion rates of mice in the experimental groups were significantly increased,and the time to the first black stool defecation was significantly shortened,indicating that Lacticaseibacillus paracasei SMR-0128 possessed good intestinemoistening and laxative functions.
