中文摘要: 为检测海胆共附生菌DNC2的抑菌活性组分,并探究抑菌活性组分的抗肿瘤作用,将海胆共附生菌DNC2的菌体和发酵液过0.22 μm滤膜,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)检测硫酸铵沉淀的发酵液蛋白组分,采用滤纸片法检测海胆共附生菌DNC2菌体、去菌体发酵液及发酵液蛋白组分的抑菌活性,琼脂块法测定抑菌蛋白组分对真菌的抑制活性,并采用海虾毒性法和噻唑蓝(methyl thiazolyl tetrazolium,MTT)法评价抑菌蛋白的抗肿瘤活性。结果表明,海胆共附生菌DNC2发酵液的抑菌活性优于菌体,菌株发酵液中的蛋白抑菌活性很强。海胆共附生菌DNC2胞外粗蛋白对雪腐格氏霉菌、串珠镰刀菌、西瓜尖镰孢菌、青枯雷尔氏菌、番茄灰霉菌、尖孢镰刀菌6 种植物病原指示菌具有抑制作用,其中对后4 种菌作用效果尤其明显。该活性粗蛋白还对慢性粒白血病癌细胞K562、肝肿瘤细胞HepG2、乳腺癌细胞MCF-7和乳腺癌细胞MDA-MB-231具有抑制作用,在蛋白质浓度为300 μg/mL时抑制率分别高达95.87%、90.40%、77.71%和95.67%。综上,海胆共附生菌DNC2胞外蛋白具有广谱抑菌和抗肿瘤活性。
Abstract:To identify the antimicrobial components produced by the symbiotic or epiphyte bacterium DNC2 from Strongylocentrotus intermedius and investigate their anti-tumor potential,both bacterial cells and fermentation broth of DNC2 were filtered through a 0.22 μm filter membrane,while the proteins from the ammonium sulfate-precipitated fermentation broth were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Subsequently,the antimicrobial activities of the bacterial cells,cell-free fermentation broth,and protein fractions were assessed using the filtering paper assay. Additionally,the antifungal activity of the antimicrobial components was determined using the agar block method,while the anti-tumor activity was evaluated using the prawn toxicity test and methyl thiazolyl tetrazolium (MTT) assay. The results indicated that the fermentation broth of DNC2 exhibited stronger antimicrobial activity than the bacterial cells,while the protein fraction from the fermentation broth showed potent antimicrobial effects. The extracellular crude proteins of DNC2 showed inhibitory activity against Gerlachia nivalis,Fusarium moniliforme,Fusarium axysporumf. sp. Niveum,Ralstonia solanacearum,Botrytis cinerea,and Fusarium oxysporum Schlecht,with particularly notable inhibition against the latter four pathogens. Moreover,the crude proteins also suppressed the proliferation of chronic myeloid leukemia cells K562,hepatocellular carcinoma cells HepG2,and breast cancer cells MCF-7 and MDA-MB-231. At a concentration of 300 μg/mL,the inhibition rates reached 95.87%,90.40%,77.71%,and 95.67%,respectively. In summary,the extracellular proteins of the symbiotic or epiphyte bacterium DNC2 from S. intermedius have broad-spectrum antimicrobial and anti-tumor activities.
