鮟鱇鱼骨胶原蛋白肽提取工艺优化及其体外抗氧化活性

中文摘要: 该试验以鮟鱇鱼副产物为原料提取胶原蛋白,并通过紫外光谱、傅里叶变换红外光谱扫描以及聚丙烯酰胺凝胶电泳分析其理化性质,通过单因素以及正交试验确定胶原蛋白酶解得到鮟鱇鱼骨胶原蛋白肽(Lophius litulon bone collagen peptide,LBCP)的最佳工艺制备,并测定体外DPPH 自由基、ABTS+自由基以及超氧阴离子自由基清除能力,以及对体外抗氧化能力进行评价。结果显示,鮟鱇鱼骨胶原蛋白最大紫外吸收在233 nm 处,在红外光谱中含有酰胺A、B、Ⅰ、Ⅱ、Ⅲ5 个特征吸收峰,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDS-PAGE)显示存在两条α1 以及一条α2 肽链,符合Ⅰ型胶原蛋白的特征。鮟鱇鱼骨胶原蛋白肽最佳酶解工艺条件为提取温度30 ℃、加酶量5%、酶解时间2 h 以及固液比20∶1(mg/mL),经此工艺得到的鮟鱇鱼骨胶原蛋白肽具有较好的抗氧化活性,DPPH 自由基、ABTS+自由基以及超氧阴离子自由基清除率半数抑制浓度分别为14.74、1.284、3.163 mg/mL,与酶解前相比,其抗氧化活性显著提升。
Abstract:This experiment extracted collagen from the by-products of anglerfish (Lophius litulon),and analyzed its physicochemical properties through ultraviolet (UV) spectroscopy,Fourier transform infrared spectroscopy (FT-IR),and polyacrylamide gel electrophoresis. The optimal process for collagen enzymatic hydrolysis was determined through single-factor and orthogonal experiments to obtain L. litulon bone collagen peptide(LBCP),and the in vitro DPPH free radical,ABTS+ free radical,and superoxide anion free radical scavenging capacities were measured to preliminarily evaluate its in vitro antioxidant capacity. The results showed that the maximum UV absorption of L. litulon bone collagen was at 233 nm,and there were five characteristic absorption peaks corresponding to amide A,B,I,II,and III in the infrared spectrum.Sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE)analysis showed the presence of two α1 and one α2 peptide chains,confirming its compliance with the characteristics of type I collagen. The optimal enzymatic hydrolysis process for L. litulon bone collagen peptide was identified as follows:extraction temperature of 30 ℃,enzyme dosage of 5%,enzymatic hydrolysis time of 2 h,and solid-liquid ratio of 20∶1 (mg/mL). The L. litulon bone collagen peptide obtained through this process demonstrated good antioxidant activity. The results showed that median inhibition concentrations (IC??) for DPPH free radicals,ABTS+ free radicals,and superoxide anion free radical scavenging rates were 14.74,1.284,and 3.163 mg/mL,respectively. Compared to the pre-hydrolysis state,the antioxidant activities were significantly improved.
