中文摘要: 为研究梅花鲨鱼皮类肝素(heparinoid from Halaelurus burgeri,Hep-hb)对高脂饮食诱导的胰岛素抵抗模型小鼠的肾脏保护作用,通过高脂饮食喂养诱导胰岛素抵抗小鼠模型。造模的同时进行干预,灌胃不同剂量Hep-hb 14 周,处死前检测空腹血糖并收集尿液,检测尿液中葡萄糖、微量白蛋白(microalbuminuria,mAlb)水平、总蛋白水平、尿素氮(urea nitrogen,UN)水平、尿酸(uric acid,UA)水平、肌酐(creatinine,Cr)水平和β-N-乙酰葡萄糖苷酶(N-acetylβ-glucosaminidase,NAG)水平等代谢相关指标,处死后摘取肾脏观察组织显微结构。结果显示,Hep-hb 可显著降低患病小鼠的葡萄糖、mAlb、总蛋白、UN、UA、Cr 和NAG 水平(p<0.05),改善肾脏组织的显微结构。综上,Hep-hb 可显著改善胰岛素抵抗小鼠的肾脏功能。
Abstract:This study aims to investigate the renal protective effect of sharkskin heparinoid from Halaelurus burgeri (Hep-hb) on insulin-resistant mice induced by a high-fat diet. An insulin-resistant mouse model was established through high-fat diet feeding,with simultaneous intervention involving intragastric administration of different doses of Hep-hb for 14 weeks. Before sacrifice,fasting blood glucose was measured,and urine samples were collected for the measurement of metabolism-related indicators,including glucose,microalbuminuria (mAlb),total protein,urea nitrogen (UN),uric acid (UA),creatinine (Cr),and N-acetyl-β-glucosaminidase (NAG). After sacrifice,the kidneys were excised to observe their tissue microstructure. The results showed that Hep-hb significantly reduced urinary excretion rates of glucose,mAlb,total protein,UN,UA,Cr,and NAG in the diseased mice (p<0.05) and improved the renal tissue microstructure. In conclusion,Hep-hb can significantly enhance renal function in insulin-resistant mice.
