中文摘要: 探索双水相二次萃取结合壳聚糖絮凝沉淀法分离纯化鲍鱼内脏(皱纹盘鲍)中β-葡萄糖苷酶,考察双水相体系构成、壳聚糖添加量、复合时pH值、振荡时间对分离纯化效果的影响,最终成功构建一条双水相二次萃取结合壳聚糖沉淀的高效分离纯化技术路线。在双水相第一次萃取中,体系总质量为10 g,pH自然,双水相最优体系组成为聚乙二醇600/(NH4)2SO4,其质量分数分别为18%和14%,纯化倍数为5.8;取第一次萃取上相5 g,加入NaH2PO4粉末至二次萃取总体系的17%(质量分数),形成第二次萃取新体系,纯化倍数为7.9。壳聚糖沉淀法最佳条件参数为壳聚糖添加浓度1.5 mg/mL、复合时pH5.8、振荡时间30 min。经上述工艺路线能获得酶活回收率和纯化倍数均更高的β-葡萄糖苷酶,酶比活为8.4 U/mg,总纯化倍数为42.2。
Abstract:This paper explored the separation and purification of β-glucosidase from abalone viscera by twotime aqueous two-phase extraction combined with chitosan flocculation and precipitation and investigated the influence of factors on purification efficiency,such as aqueous two-phase composition,chitosan addition,pH during complexation,and shaking time. Finally,a highly efficient separation and purification technology combining aqueous two-phase extraction with chitosan precipitation was successfully constructed. In the primary aqueous two-phase extraction process,the total system was 10 g with natural pH conditions,and the optimal composition was PEG600/(NH4)2SO4,with mass fractions of 18% and 14%. The purification factor was 5.8. By taking the upper phase 5 g of the primary extraction system and adding NaH2PO4 powder to 17% (mass fraction) of the total extraction system for secondary extraction,the secondary aqueous two-phase extraction system was developed,with a total purification factor of 7.9. The optimal conditions for the chitosan precipitation method were chitosan addition of 1.5 mg/mL,pH of 5.8 during complexation,and a shaking time of 30 min.Through the above process,β-glucosidase with higher enzyme activity recovery and purification factor was obtained,with the specific activity of 8.4 U/mg and the purification factor of 42.2.
