不同比色法测定枸杞中总黄酮含量及其抗氧化活性比较

中文摘要: 采用硼氢化钠/氯醌(sodium borohydride /chloranil,SBC)比色法、NaNO2-Al(NO3)3 比色法和AlCl3 比色法测定枸杞总黄酮含量,并对其黄酮提取物的体外抗氧化活性进行对比分析。结果表明:SBC 比色法的精密度、稳定性、重现性和加标回收率均良好,且综合表现优于其他两种方法;标准品的选择对测定结果存在影响,以芦丁或者槲皮素为标准品能够相对准确地反映枸杞总黄酮含量;以槲皮素为标准品时,SBC 比色法所测得枸杞中总黄酮含量为10.48~116.97 mg QE/g,是NaNO2-Al(NO3)3 比色法所测得含量的4~31 倍,AlCl3 比色法测得的含量相对较低;不同枸杞部位的总黄酮含量差异较大,黑果枸杞(干)和红果枸杞根(干)的总黄酮含量最高,且二者差异不显著,SBC 比色法所测得红果枸杞鲜果和干果的总黄酮含量分别为10.48 mg QE/g 和72.22 mg QE/g;体外抗氧化活性结果显示,黑果枸杞(干)和红果枸杞根(干)的黄酮提取物对DPPH 自由基和ABTS 阳离子自由基的清除率最高。总之,SBC 比色法能够检测的黄酮类化合物种类更全面,所测得的枸杞中黄酮含量结果更准确;红果枸杞根(干)和黑果枸杞(干)中的黄酮类化合物较高,其体外抗氧化活性也较高。
Abstract:The total flavonoid content of Lycium was determined using sodium borohydride/chloranil (SBC)colorimetry, NaNO2-Al(NO3)3 colorimetry, and AlCl3 colorimetry. In addition, the in vitro antioxidant activity of the flavonoid extracts was compared. The results showed that SBC colorimetry exhibited good precision, stability, repeatability, and recovery rates, and its overall performance was superior to the other two methods.The choice of standard affected the measurement results. Using rutin or quercetin as the standard provided a relatively accurate reflection of the total flavonoid content in Lycium. When quercetin was used as the standard, SBC colorimetry measured the total flavonoid content in Lycium as 10.48-116.97 mg QE/g, which was 4-31 times higher than the content measured by NaNO2-Al(NO3)3 colorimetry. The content measured by AlCl3 colorimetry was relatively lower. The total flavonoid content varied significantly among different parts of Lycium. The highest content was found in the dried root of L. barbarum L. and dried L. ruthenicum, with no significant difference between the two. The total flavonoid content in fresh and dried fruit of L. barbarum L., measured by SBC colorimetry, was 10.48 mg QE/g and 72.22 mg QE/g, respectively. In terms of in vitro antioxidant activity, the flavonoid extracts from dried L. ruthenicum and the dried root of L. barbarum L. exhibited the highest DPPH and ABTS+ radical scavenging activities. In conclusion, SBC colorimetry detected a broader range of flavonoid compounds and provided more accurate measurements of total flavonoid content in Lycium.The flavonoid content and antioxidant activity in the dried root of L. barbarum L. and dried L. ruthenicum were higher than in other samples.
