中文摘要: 该文建立微量双波长法测定直链淀粉与支链淀粉含量的方法,并用此法测定15 种马铃薯的直链淀粉和支链淀粉的含量。结果表明,直链淀粉和支链淀粉80 ℃水浴25 min 分散效果好,标准曲线最佳显色条件为直链淀粉和支链淀粉混合标准溶液12 μL、乙酸溶液23 μL、0.2% 碘化钾试剂10 μL、蒸馏水205 μL,直链淀粉与支链淀粉的测定波长分别为619、534 nm,参比波长分别为451、751 nm,在10~60 min,ΔA 直和 ΔA 支基本稳定,直链淀粉与支链淀粉含量误差值均不超过3%,相对标准偏差均小于1%,直链淀粉、支链淀粉回收率分别为 98.26%~99.50%、99.32%~100.07%,此方法测定的15 种马铃薯中的直链淀粉和支链淀粉含量分别为19.46%~24.93%、62.12%~78.88%。
Abstract:A micro dual-wavelength method for determining the content of amylose and amylopectin was established. This method was then used to determine the content of amylose and amylopectin in 15 potato varieties.The results showed that amylose and amylopectin demonstrated ideal dispersion effects after being placed in a water bath at 80 ℃ for 25 min. The optimal color reaction conditions for the standard curve were 12 μL mixed standard solution of amylose and amylopectin, 23 μL acetic acid solution, 10 μL 0.2% iodine reagent, and 205 μL distilled water. The determination wavelengths for amylose and amylopectin were 619 nm and 534 nm,respectively, and the reference wavelengths were 451 nm and 751 nm, respectively. The ΔAamylose and ΔAamylopectin were basically stable within 10-60 min. The error values of amylose and amylopectin content did not exceed 3%, and the relative standard deviations were both <1%. The recovery rates of amylose and amylopectin were 98.26%-99.50% and 99.32%-100.07%, respectively. The content of amylose and amylopectin in the 15 potato varieties determined by this method was 19.46%-24.93% and 62.12%-78.88%, respectively.
