中文摘要: 三峡库区红阳猕猴桃资源丰富,为高值化利用红阳猕猴桃,该文对其多糖成分进行研究。通过超声波辅助水提醇沉法提取红阳猕猴桃中的多糖成分,采用DEAE-52 纤维素柱层析对红阳猕猴桃粗多糖(Actinidia chinensis cv.Hongyang,ACP)进行分离,综合考察得率、抗氧化活性等,筛选出主要多糖组分,进一步通过SephadexG-100 凝胶柱层析得到高纯度多糖组分;对ACP 和高纯度多糖组分进行结构表征,并对比两者的基本化学成分及抗氧化活性。结果显示,三峡库区红阳猕猴桃经超声波辅助水提醇沉法提取的粗多糖得率为(16.23±0.38)%;通过柱层析纯化得到洗脱组分ACP-3-G,其多糖、糖醛酸和还原糖含量均高于ACP;与ACP 对比,ACP-3-G 具有三螺旋结构,含有糖醛酸、α型和β 型糖苷键构型及吡喃环构型,呈片状及纤维丝状,具有良好的溶解性;ACP 对DPPH 自由基、ABTS+自由基清除率及总还原力均高于ACP-3-G,ACP-3-G 对羟基自由基的清除率略高于ACP。研究表明,红阳猕猴桃多糖具有一定抗氧化活性。
Abstract:Actinidia chinensis cv. Hongyang has rich resources in the Three Gorges Reservoir area. The polysaccharide components of Actinidia chinensis cv. Hongyang were studied for its high-value utilization. Polysaccharides were extracted from Actinidia chinensis cv. Hongyang by adopting the ultrasonic-assisted water extraction and alcohol precipitation method. Meanwhile, DEAE-52 cellulose column chromatography was employed to separate the crude polysaccharide of Actinidia chinensis cv. Hongyang (ACP). Based on the thorough investigation of the yield and antioxidant activity the main polysaccharide components were screened, and the highpurity polysaccharides were obtained by SephadexG-100 gel column chromatography. Meanwhile, structural characterization was conducted on ACP and the high-purity polysaccharide content, with their basic chemical components and antioxidant activity compared. The results showed that the yield of crude polysaccharides from Actinidia chinensis cv. Hongyang was (16.23±0.38)% by adopting ultrasonic-assisted water extraction and alcohol precipitation. The elution component ACP-3-G was obtained by column chromatography purification, and its polysaccharide, uronic acid, and reducing sugar contents were higher than those of ACP. Compared with ACP, ACP-3-G had a triple helix structure, containing uronic acid, α and β glucosidic bond configurations and pyranoid ring configurations, which were flaky and fibrous with sound solubility. The scavenging rate and reducing power of ACP for DPPH and ABTS+ free radicals were higher than those of ACP-3-G, and the scavenging rate of ACP-3-G for hydroxyl free radicals was slightly higher than that of ACP. The results showed that the polysaccharides of Actinidia chinensis cv. Hongyang had certain antioxidant activity.
