中文摘要: 以灵芝加工后的菌柄为原料,研究其多糖、三萜的提取工艺及抗肿瘤、抗氧化活性。采用超声波-酶促辅助提取法优化加工废料多糖、三萜提取工艺;采用3- (4,5-二甲基-2-噻唑基)-2,5-二苯基四氮唑溴化物[3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide,MTT]法研究灵芝废弃物中三萜和多糖对两种癌细胞的抑制作用,并对多糖进行红外光谱表征;采用体外抗氧化模型对三萜和多糖进行活性研究。结果表明,松杉灵芝菌柄中多糖及三萜得率的最佳提取工艺条件为乙醇浓度40%、超声时间30 min、提取温度50 ℃、固液比1∶25 (g/mL)、酶添加量500 U/g。在此条件下,多糖、三萜得率分别为(1.570±0.026)%、(1.635±0.020)%;多糖和三萜浓度与肝癌细胞(HepG2)的抑制率呈正相关,当多糖浓度达到1 000 μg/mL 时,对HepG2 和Caki-1 两种肿瘤细胞的抑制率分别为(81.73±2.20)%、(71.39±2.73)%;当三萜浓度为500 μg/mL 时,对两种肿瘤细胞的抑制率分别为(64.33±1.16)%、(76.95±2.20)%;红外光谱结果显示该多糖为β-吡喃糖;体外抗氧化结果表明,该三萜、多糖均具有良好的抗氧化活性。
Abstract:The extraction of polysaccharide and triterpenoids from the stipes (processing residues) of Ganoderma tsugae Murr was explored. Furthermore, the antitumor and antioxidant activities of the extracted polysaccharide and triterpenoids were determined. The ultrasound-assisted enzymatic method was used for extraction. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) method was employed to study the inhibitory effects of the extracted triterpenoids and polysaccharide on two cancer cell lines (HepG2 and Caki-1). The polysaccharide was characterized by infrared spectroscopy(IR). Furthermore, the antioxidant activities of triterpenoids and polysaccharide in vitro were determined. The results showed that the optimal extraction conditions were ethanol concentration of 40%, ultrasonic time of 30 min, ultrasonic temperature of 50 ℃, solid-liquid ratio of 1∶25 (g/mL), enzyme dosage of 500 U/g. Under these conditions, the yields of polysaccharide and triterpenoids were (1.570±0.026)% and (1.635±0.020)%, respectively. The concentration of polysaccharide and triterpenoids was proportional to the inhibition rate on HepG2 cells. At the concentration of 1 000 μg/mL, polysaccharide showed the inhibition rates of (81.73±2.20)% and (71.39±2.73)% on HepG2 and Caki-1 cells, respectively. At the concentration of 500 μg/mL, triterpenoids demonstrated the inhibition rates of (64.33±1.16)% and (76.95±2.20)% on HepG2 and Caki-1 cells, respectively. The IR showed that polysaccharide was β-pyranose. Both the extracted polysaccharide and triterpenoids showcased strong antioxidant activities in vitro.
