中文摘要: 为建立一种高效便捷的黄曲霉毒素B1(aflatoxin B1,AFB1)检测方法,该研究以硫代乙酰胆碱(acetylcholine thio,ATCh)为底物,探究AFB1 对乙酰胆碱酯酶(acetylcholinesterase,AChE)生物活性情况的影响。结果表明,硫代胆碱(thiocholine,TCh)能以Au-S 共价形式吸附在金纳米粒子(gold nanoparticles,AuNPs)表面,导致AuNPs 发生聚合,其颜色由酒红色逐渐转变为蓝灰色,同时在波长522 nm 处的局域表面等离子共振吸收峰也顺次降低。当样品中存在AFB1时,乙酰胆碱酯酶的生物活性受到抑制,减弱硫代乙酰胆碱水解生成TCh 的能力。基于测定抑制前后522 nm处的紫外-可见吸收峰值变化,该研究构建一种快速、简便、灵敏的AFB1 可视化比色方法,并实现了样品中痕量AFB1 的检测。最优试验条件为试验介质pH7.4,乙酰胆碱酯酶浓度2 500 mU/mL,底物浓度5 μmol/L,反应时间60 min。AFB1 检测的线性范围为0.1~20 ng/mL,检出限达到0.03 ng/mL,实际花生油样品的加标回收率为93.4%~104.9%,综上,该方法具有较高的灵敏度与准确性。
Abstract:An efficient and convenient method for determining aflatoxin B1 (AFB1) was established. With acetylcholine thio (ATCh) as the substrate, the effect of AFB1 on the bioactivity of acetylcholinesterase (AChE)was studied. The results indicated that thiocholine(TCh) could be adsorbed on the surface of gold nanoparticles (AuNPs) in the form of Au-S covalent form, which led to the polymerization of AuNPs and the gradual color change from burgundy to blue-gray in visible light. Meanwhile, the local surface plasmon resonance absorption peaks at the wavelength of 522 nm decreased in turn. When AFB1 was present in a sample, the biological activity of AChE was inhibited, which reduced hydrolysis of ATCh to TCh. Based on the determination of the change of UV-vis absorption peaks at 522 nm before and after the inhibition, a rapid, simple, and sensitive colorimetric method for the visualization of AFB1 was constructed, which realized and the detection of trace AFB1 in samples. The optimal reaction conditions were as follows: pH7.4, AChE concentration of 2 500 mU/mL, substrate concentration of 5 μmol/L, and reaction time of 60 min. The linear range of AFB1 was 0.1-20 ng/mL, and the limit of detection was 0.03 ng/mL. The spiked recovery of the actual peanut oil samples ranged from 93.4% to 104.9%. The results indicated that the established method had high sensitivity and accuracy for the determination of AFB1 in food.
