产蛋白酶海洋芽孢杆菌的分离鉴定及鲬鱼抗氧化肽的制备

中文摘要: 蛋白酶水解鱼类加工副产物,制备抗氧化活性肽,是提高其附加值的重要途径。该文从海州湾海泥中筛选得到一株产蛋白酶菌株H-1,结合形态鉴定、生理生化反应和16S rDNA 基因序列分析鉴定为芽孢杆菌(Bacillus siamensis)。菌株H-1 产蛋白酶的最适反应温度为55 ℃、pH 值为8,具有较好的温度、pH 稳定性,在谷胱甘肽、β-巯基乙醇等还原剂中具有良好的稳定性。50 mmol/L 的金属离子Co2+、Mn2+、K+、Mg2+对酶活力有明显促进作用,Fe3+、Ba2+、Zn2+对酶活力有抑制作用。该酶水解鲬鱼加工副产物制备具有抗氧化作用的活性肽,在浓度为10 mg/mL 时,DPPH 自由基清除率为80%、羟基自由基清除率为80%、超氧阴离子自由基清除率为52.7%,还原力为0.51。结果表明,该酶可以明显提高鲬鱼副产物的应用价值。
Abstract:Preparing antioxidant peptides by digestion with protease is an important way to increase the value added of fish by-products.A protease-producing strain H-1 was isolated from the sea mud of Haizhou Bay and identified as?Bacillus?siamensis?based on the morphological,physiological,biochemical,and 16S rDNA gene sequence evidence. The optimal reaction conditions of the protease produced by strain H-1 were 55 ℃and pH8. The protease had good thermostability and pH stability and demonstrated good stability in solvents such as glutathione,β-mercaptoethanol. The protease activity was improved by 50 mmol/L Co2+,Mn2+,K+,and Mg2+,while it was inhibited by Fe3+,Ba2+,and Zn2+.This enzyme hydrolyzed the by-products of?Platycephalus indicus?to produce active peptides with antioxidant properties.At a concentration of 10 mg/mL,the antioxidant peptides showed the DPPH,hydroxyl,and superoxide anion free radical clearance rates and the reducing power of 80%,80%,52.7%,and 0.51,respectively. The results indicated that the protease significantly enhanced the application value of the by-products of?P.indicus.
