中文摘要: 探究蒲公英多糖、黄芪多糖、枸杞多糖、山药多糖与当归多糖5 种多糖对木糖葡萄球菌(Staphylococcus xylosus) TG022增殖和产酶能力的影响,进一步探究蒲公英多糖、当归多糖与木糖葡萄球菌在肠道微生物调节中的作用。分别配制以上述5 种多糖作为唯一碳源的培养基,与木糖葡萄球菌TG022共同培养,以菌株生长曲线、pH值、酶活为指标,筛选对木糖葡萄球菌TG022有增益作用的多糖;通过体外模拟肠发酵试验,探究筛选的多糖对肠道微生物的影响。结果表明,当归多糖和蒲公英多糖对木糖葡萄球菌TG022的协同增长效果最优,其生长曲线与酶活表现出协同增加的趋势,其他3 种多糖对木糖葡萄球菌TG022无协同增长效果;体外模拟肠道发酵液的16S rRNA基因高通量测序结果表明,与阴性对照组相比,添加当归多糖与蒲公英多糖后发酵的木糖葡萄球菌显著降低了致病菌属的相对丰度,增加了有益菌属相对丰度。蒲公英多糖和当归多糖能够作为唯一的碳源促进木糖葡萄球菌TG022生长,且在菌株的增殖量、酶活、体外肠道模拟发酵中协同发挥较好作用。
Abstract:The proliferation and enzyme production of Staphylococcus xylosus TG022 treated with polysaccharides from five polysaccharides (Taraxaci Herba,Astragali Radix,Lycii Fructus,Dioscoreae Rhizoma,and Angelicae Sinensis Radix) with both edible and medicinal values were studied.The regulating effects of Taraxaci Herba polysaccharide and Angelicae Sinensis Radix polysaccharide combined with S.xylosus TG022 on intestinal flora were explored.The five polysaccharides were respectively used as the sole carbon sources for the culture of S.xylosus TG022,and the polysaccharides suitable for strain growth was screened out based on the growth curve,pH,and enzyme activity.Furthermore,in vitro intestinal fermentation experiments were carried out to examine the effects of the polysaccharides screened out on intestinal flora.The results showed that Angelicae Sinensis Radix polysaccharide and Taraxaci Herba polysaccharide had the best effects on the growth of S.xylosus TG022,increasing the biomass and enzyme activity.The polysaccharides from the other three Chinese herbal medicines had no promoting effect on the growth of S.xylosus TG022.The results of 16S rRNA gene high-throughput sequencing showed that compared with the negative control,fermentation of S.xylosus TG022 with Angelicae Sinensis Radix or Taraxaci Herba polysaccharide reduced the relative abundance of pathogenic microorganisms and increased the relative abundance of beneficial microorganisms.Angelicae Sinensis Radix or Taraxaci Herba polysaccharide could be used as the only carbon source to promote the growth of S.xylosus TG022,promoting the proliferation,enzyme production,and in vitro intestinal fermentation.S.xylosus TG022 combined with each of the two polysaccharides can regulate the relative abundance of pathogenic microorganisms and beneficial microorganisms in the intestine.
