中文摘要: 为解析高、低温季节对养殖长牡蛎(Crassostrea gigas)微生物群落组成,及微生物群落变化对长牡蛎品质和食品安全的潜在影响,利用16s扩增子高通量测序技术,对高温季节和低温季节采集自青岛鳌山湾的长牡蛎软体组织微生物群落结构进行分析。高通量测序结果表明,高温季节长牡蛎样本软体组织中的细菌群落主要由变形菌门(Proteobacteria,40.35%)、厚壁菌门(Firmicutes,19.87%)、放线菌门(Actinobacteria,12.23%)和拟杆菌门(Bacteroidetes,10.39%)组成;在属水平上优势菌为双歧杆菌属(Bifidobacterium,5.11%)、栖水菌属(Enhydrobacter,5.03%)、内源性单胞菌属(Endozoicomonas,4.81%)和甲基杆菌属(Methylobacterium,3.04%)。低温季节长牡蛎样本软体组织中细菌群落发生变化,放线菌门占比大幅降低至4.14%,蓝细菌门(Cyanobacteria,9.05%)占比由2.61%上升至9.05%;在属水平上鞘氨醇单胞菌属(Sphingomonas,21.13%)、蓝细菌(Cyanobacteria,7.70%)和支原体属(Mycoplasma,5.08%)占比上升,成为优势菌,而双歧杆菌属(0.58%)、栖水菌属(0.05%)占比大幅降低;弧菌属(Vibrio)在外套膜组织中占比最高,整体占比由高温季节的1.25%降低至0.10%,与传统培养结果一致。从高温季节到低温季节,长牡蛎样品中微生物群落丰富度略有增加,而多样性有所下降。该研究可为评估不同季节长牡蛎品质与安全的影响提供参考。
Abstract:To investigate the microbiota variation of Pacific oyster Crassostrea gigas in the hot and cold seasons and its potential impact on the quality of C.gigas and food safety.The 16S rRNA gene sequencing was used to detect the composition and variation of the microbial community in the soft tissues of C.gigas during the hot and cold seasons,which were sampled at the Aoshan Bay in Qingdao,Shandong Province.The high-throughput sequencing results showed that bacteria colonized in C.gigas sampled in the hot season were dominated by Proteobacteria(40.35%),Firmicutes(19.87%),Actinobacteria(12.23%),and Bacteroidetes(10.39%).At the genus level,Bifidobacterium Spp.(5.11%),Enhydrobacter(5.03%),Endozoicomonas(4.81%),and Methy-lobacterium(3.04%)were predominant.However,in the cold season,the main changes of microbiota in the soft tissue of C.giga were that Actinobacteria dropped to 4.14%,and Cyanobacteria increased from 2.61%to 9.05%.At the genus level,the dominant bacteria were Sphingomonas(21.13%),Cyanobacteria(7.70%),and Mycoplasma(5.08%),which significantly increased compared with those in the C.gigas sampled in the hot season.In contrast,Bifidobacterium(0.58%)and Enhydrobacter(0.05%)significantly decreased.The Vibrio spp.accounted for the highest proportion(3.03%)in mantle tissue,and the overall abundance reduced from 1.25%in the hot season to 0.10%in the cold season,which was consistent with the results of the culture-based method.From hot season to cold season,the richness of microbial community in C.gigas samples increased slightly,while the diversity decreased.These results provided insights for further assessing the impact of microbiota on the quality and safety of C.gigas collected in different seasons.
