紫苏籽多糖对高脂饮食小鼠脂代谢和抗氧化作用的影响

中文摘要: 为评估紫苏籽多糖对高脂饮食(high-fat diet,HF)导致的小鼠脂代谢异常和氧化应激的改善作用。选取150只健康的昆明小鼠,随机分为对照组、高脂日粮组、低剂量紫苏籽多糖组、中剂量紫苏籽多糖组和高剂量紫苏籽多糖组,试验共8周。结果表明:试验结束后,高脂日粮组小鼠体质量显著高于对照组小鼠。日粮中紫苏籽多糖的添加显著降低了HF饮食小鼠肝脏指数以及血清中谷草转氨酶和谷丙转氨酶、甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、低密度脂蛋白和血糖水平,以及显著提高了小鼠血清中高密度脂蛋白水平。此外,紫苏籽多糖的添加也显著降低了小鼠肝脏中TC和TG的含量,提高了肝脏和肠道中超氧化物歧化酶、过氧化物酶和谷胱甘肽过氧化物酶的活性,肝脏和肠道中丙二醛水平也显著降低。mRNA和蛋白免疫印记结果表明紫苏籽多糖显著降低了HF饮食小鼠肝脏脂肪酸合成酶的mRNA和蛋白表达水平,显著提高了肉碱棕榈酰转移酶和脂肪组织甘油三酯水解酶的mRNA和蛋白表达水平。综上所述,紫苏籽多糖缓解了HF饮食导致的小鼠氧化应激和脂代谢异常。

Abstract:The aim of this study was to evaluate the ameliorating effects of Perilla frutescens seeds polysaccharide on abnormal lipid metabolism and oxidative stress in mice treated with a high-fat diet.One hundred and fifty healthy Kunming mice were selected and randomly divided into control,high-fat(HF)diet,low-dose Perilla frutescens seeds polysaccharide(L-PFSP),medium-dose Perilla frutescens seeds polysaccharide(M-PFSP),and high-dose perilla frutescens seeds polysaccharide(H-PFSP)groups.The results showed that,at the end of the 8-week experimental period,mice in the HF diet group weighed significantly more than those in the control group.Dietary supplementation with Perilla frutescens seeds polysaccharide(M-PFSP and H-PFSP groups)significantly decreased liver index values and serum aspartate aminotransferase (AST),alanine aminotransferase,triglyceride(TG),total cholesterol(TC),low-density lipoprotein-C,and glucose levels in mice fed an HF diet,and significantly increased serum high-density lipoprotein-C levels.Moreover,the addition of Perilla frutescens seeds polysaccharide(M-PFSP and H-PFSP groups)significantly reduced TC and TG levels in the liver,and increased the levels of superoxide dismutase and peroxidase in the liver and intestine.Catalase and glutathione peroxidase activities and malondialdehyde levels in the liver and intestine were also significantly reduced.The results of mRNA and western blotting analyses showed that Perilla frutescens seeds polysaccharide significantly reduced the mRNA and protein expression levels of fatty acid synthetase in the liver of mice fed an HF diet,and significantly increased carnitine palmitoyltransferase I and adipose triglyceride lipase mRNA and protein expression levels.In conclusion,Perilla frutescens seeds polysaccharide alleviated oxidative stress and abnormal lipid metabolism in mice fed an HF diet.
