中文摘要: 以西藏昌都地区9个不同县出产的芜菁为对象,研究其总黄酮和总皂苷的含量、抗氧化活性及对过氧化氢诱导RAW264.7细胞氧化应激损伤的保护作用。研究结果表明,9个不同县出产的芜菁中总黄酮、总皂苷的含量存在差异。其中贡觉县芜菁中总黄酮含量最高,江达县的含量最低;贡觉县芜菁中总皂苷含量最高,边坝县的含量最低。体外DPPH自由基及ABTS+自由基清除活性试验结果表明,贡觉县、丁青县和八宿县芜菁样品的抗氧化活性优于江达县和边坝县。构建过氧化氢诱导RAW264.7细胞氧化应激损伤模型,试验结果表明贡觉县、八宿县、类乌齐县的芜菁样品能够促进氧化应激RAW264.7细胞模型中细胞的增殖,并清除自由基,降低细胞内活性氧的水平,缓解氧化应激对细胞所造成的损伤,改善细胞对外界氧化应激刺激的调控能力。综上,不同地区芜菁中总黄酮和皂苷的含量以及抗氧化活性具有差异,为西藏地区芜菁的开发及利用提供了一定的科学依据。
Abstract:Turnips produced in 9 different counties of Qamdo area in Tibet were studied.The contents of total flavonoids and total saponins,antioxidant activity and protective effects on oxidative stress induced by hydrogen peroxide in RAW264.7 cells were explored.The results showed that there were differences in the content of total flavonoids and total saponins between turnips from different counties.Among them,the total flavonoids content of Gongjue County was the highest,and Jiangda County was the lowest;the total saponin content of Guanzhi County was the highest,and Bianba County was the lowest.The results of DPPH and ABTS+scavenging free radical activity in vitro showed that the antioxidant activities of Gongjue County,Dingqing County and Basu County were superior to those of Jiangda County and Bianba County.By establishing oxidative stress injury model induced by hydrogen peroxide in RAW264.7 cells,the results showed that the turnips samples from Gongjue County,Basu County and Wuqi County could promote cell proliferation and scavenge free radicals in the oxidative stress model.They could also reduce the level of intracellular reactive oxygen species,alleviate the damage caused by oxidative stress on cells,and improve the cell's ability to regulate external oxidative stress.In conclusion,the content of total flavonoids and saponins,and the antioxidant activities in turnips from different regions were different,which provided a scientific basis for the development and utilization of turnips in Tibet.
