中文摘要: 基于基质辅助激光解吸电离-飞行时间质谱(MALDI-TOFMS)技术建立一种羊乳中磷酸化肽段的检测方法。以商品化二氧化钛(TiO2)为富集材料用于羊乳中低丰度磷酸肽的富集,并优化了商品化TiO2富集羊乳中磷酸肽的最佳吸附洗脱条件,得到的结果为,孵育缓冲液为乙腈∶水(50∶50,体积比)+1%三氟乙酸,洗脱液为氨水15%。接着探讨了脱脂对检测结果的影响,结果表明,通过MALDI-TOF MS分析实际乳磷酸肽时,需要对乳类样品进行脱脂才能更完整的鉴定磷酸肽,从羊乳中鉴定出八个磷酸化肽段通过Mascot数据库检索。本研究为乳类磷酸肽指纹图谱的建立提供了理论基础,指纹图谱的建立能够应用于掺假乳的快速鉴定。

Abstract:A novel determination method was developed for the phosphopeptide of goat milk which based on MALDI-TOF MS technology.Primarily,commercial TiO2was recommended for the process of phosphopeptide enrichment,and the optimized resultofenrichmentprocess suggests thatthe content ofphosphopeptide reached a maximum by using acetonitrile∶water(50∶50,volume ratio)+1%trifluoroacetic acid as the incubation buffer while using 15%ammonia as eluent.Subsequently,the effect of remove fat from milk for the detection resultwas discussed,and the resultItconfirmed thatitis necessary to remove fat from milk before make a more complete identification ofthe phosphopeptides in milk samples.MALDI-TOF MS was used to determine the enriched phosphopeptide of goat milk.Through fingerprints analysis and comparison with the Mascot database,eight phosphopeptides have been identified in goatmilk.The present study provides the theory base for the establishmentofthe dairy phosphopeptide fingerprintand contributes for rapid identification ofadulterated milk.
