中文摘要: 为研究广东虫草子实体多糖的体外抗氧化活性和体液免疫调节功能,该研究采用热水浸提醇沉法制备广东虫草子实体粗多糖(CGFCP),经二乙氨基乙基(diethylaminoethyl,DEAE)纤维素DE-52 凝胶纯化获得两种纯化多糖(CGFPP-W 和CGFPP-S);采用气相色谱-质谱(gas chromatography-mass spectrometry,GC-MS)方法检测多糖的单糖组成,测定其体外抗氧化活性,并通过小鼠模型评估CGFCP 低、中、高剂量对抗体生成细胞及血清溶血素的影响。结果表明,CGFCP、CGFPP-W 和CGFPP-S 的单糖主要由葡萄糖、甘露糖、阿拉糖和半乳糖组成;随着浓度增加,三者均表现出较强的体外抗氧化活性,且CGFCP 在浓度为10 mg/mL 时对清除羟基自由基、DPPH 自由基及总还原力方面的抗氧化活性强于纯化多糖(CGFPP-W 和CGFPP-S);低、中、高剂量的CGFCP 均可提升小鼠溶血空斑数及抗体体积数,其中高剂量组体液免疫调节能力显著高于阴性对照组。综上,广东虫草子实体多糖具有一定的抗氧化活性和体液免疫调节功能。
Abstract:To investigate the in vitro antioxidant activity and humoral immunomodulatory function of Cordyceps guangdongensis fruiting body polysaccharides,crude polysaccharides of C. guangdongensis fruiting body(CGFCP) were prepared by hot water extraction and ethanol precipitation in this study. Two purified polysaccharides (CGFPP-W and CGFPP-S) were obtained through subsequent purification with diethylaminoethyl(DEAE) cellulose DE-52 gel. The monosaccharide composition of the polysaccharides was determined by gas chromatography-mass spectrometry (GC-MS),and their in vitro antioxidant activity was assessed. A mouse model was used to evaluate the effects of low,medium,and high doses of CGFCP on antibody-producing cells and serum hemolysin. The results showed that the monosaccharide composition of CGFCP,CGFPP-W,and CGFPP-S was primarily glucose,mannose,arabinose,and galactose. With increasing concentration,all three polysaccharides exhibited strong in vitro antioxidant capacity. Compared to the purified CGFPP-W and CGFPPS,the CGFCP at 10 mg/mL demonstrated stronger antioxidant activity (hydroxyl and DPPH free radical scavenging ability,as well as total reducing power). Low,medium,and high doses of CGFCP increased the number of hemolytic plaques and titer in mice,with the high-dose group showing a significantly stronger humoral immunomodulatory capacity than the negative control group. In conclusion,C. guangdongensis fruiting body polysac-charides possess certain antioxidant activity and humoral immunomodulatory function.
