中文摘要: 以前期筛选的产酸菌和产酯化酶菌株构建二元混菌发酵体系。以体系的OD600 nm为指标确立无竞争抑制作用的二元混菌发酵体系。利用气相色谱质谱联用仪(gas chromatography-mass spectrometry,GC-MS)测定无竞争抑制作用的二元混菌发酵体系的发酵液中脂肪酸乙酯等风味物质。结果显示,以产酯化酶突变株(Sphingomonas paucimobilis) ARUV3-26与丁酸梭菌(Clostridium butyricum) GD1-1构建的体系中的脂肪酸乙酯含量最高,其中乙酸乙酯1.44 g/L、丁酸乙酯1.32 g/L、乳酸乙酯1.17 g/L和己酸乙酯1.04 g/L。通过单因素和响应面法优化二元混菌发酵体系的发酵条件,发现二元混菌发酵体系中产酯化酶菌株的接种时间为 第3.4天、产酯化酶菌株接种量为5.3%、二元混菌发酵时间为5.3 d,二元混菌发酵体系酯类物质及含量为乙酸乙酯(0.87±0.02) g/L、丁酸乙酯(0.91±0.03) g/L、乳酸乙酯(1.16±0.05) g/L和己酸乙酯(1.48±0.11) g/L。该研究的二元混菌体系易于构建,发酵液中风味物质丰富,酯化反应不需要辅因子,可保证产品的风味和质量。
Abstract:A fermentation system with binary mixed bacteria was constructed with the acid-producing bacteria and the esterase-producing strains screened in the preliminary study. The competitive inhibition of the system was evaluated based on OD600 nm. The fatty acid ethyl esters and other flavor substances in the system without competitive inhibition were quantified by gas chromatography-mass spectrometry. The results showed that the yield of fatty acid ethyl esters was the highest in the fermentation system consisting of Sphingomonas paucimobilis ARUV3-26 and Clostridium butyricum GD1-1,including ethyl acetate at 1.44 g/L,ethyl butyrate at 1.32 g/L,ethyl lactate at 1.17 g/L,and ethyl hexanoate at 1.04 g/L. The fermentation conditions of the system were optimized by single factor and response surface methods. The optimized conditions were as follows:inoculation of the esterase-producing strain at the time point of 3.4 d and an amount of 5.3% and the fermentation time of 5.3 d. Under these conditions,the yields of ethyl acetate,ethyl butyrate,ethyl lactate,and ethyl hexanoate produced by the system were (0.87±0.02),(0.91±0.03),(1.16±0.05),and(1.48±0.11) g/L,respectively. The fermentation system with binary mixed bacteria was characterized by easy construction,fermentation broth rich in flavor substances,and esterification without the need for cofactors,thus ensuring the flavor and quality of the product.
