中文摘要: 该研究旨在初步探究屎肠球菌(Enterococcus faecium R-026)和枯草芽孢杆菌(Bacillus subtilis R-179)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)诱导的帕金森氏病小鼠的作用,着重探讨其对小鼠肠道菌群的影响。通过腹腔注射MPTP建立帕金森(Parkinson′s disease,PD)小鼠模型,在给予屎肠球菌联合枯草芽孢杆菌(B. subtilis R-179 and E. faecium R-026,BE)后,测定小鼠血清相关指标,16S rDNA鉴定高通量测序分析技术检测各组小鼠肠道菌群丰度变化。行为学结果表明,与模型组(MPTP组)相比,益生菌治疗组(BE组)小鼠爬杆时间变短,悬挂实验得分有所升高,旷场实验中5 min运动总路程显著增加。生化指标结果显示,BE组较MPTP组小鼠血清中的超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活性升高,谷胱甘肽(glutathione,GSH)含量升高,丙二醛(malondialdehyde,MDA)含量下降,脑组织中多巴胺(dopamine,DA)含量升高。蛋白质免疫印迹法结果表明,经益生菌治疗后,BE组小鼠脑组织中血红素氧合酶1(hemeoxygenase-1,HO-1)、转录因子NF-E2相关因子2(nuclear factor erythroid 2-related factor 2,Nrf2)蛋白表达水平升高。16S rDNA结果表明,与空白组(Con组)小鼠相比,Proteobacteria、Bacteroidetes和Verrucomicrobia在 PD小鼠中的丰度明显增加,而Actinobacteria、Patescibacteria、Campylobacterota、Cyanobacteria和 Deferribacteres的丰度明显降低。而益生菌干预可以纠正上述PD小鼠肠道菌群门水平的变化。屎肠球菌联合枯草芽孢杆菌对MPTP诱导的PD小鼠有显著的治疗作用,其机制可能与抗氧化作用、肠道菌群间的共同作用有关。

Abstract:This study aims to preliminarily explore the effects of Enterococcus faecium R-026 and Bacillus subtilis R-179 on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced Parkinson′s disease(PD)mice,with a focus on their impact on the intestinal microbiota of mice. PD mouse models were established by intraperitoneal injection of MPTP. After administration of a combination of E. faecium and B. subtilis (B. subtilis R-179 and E. faecium R-026,BE),serum-related indicators were measured,and high-throughput sequencing analysis of 16S rDNA was used to detect changes in the abundance of intestinal microbiota in each group of mice. Behavioral results showed that compared with the model group (MPTP group),the BE group had shorter pole-climbing time,higher hanging test scores,and a significantly increased total distance traveled in the open field test within 5 minutes. Biochemical index results indicated that the BE group had higher activities of superoxide dismutase (SOD) and catalase (CAT),higher glutathione (GSH) content,lower malondialdehyde (MDA)content in serum,and higher dopamine (DA) content in brain tissue compared with the MPTP group. Western blot results showed that after probiotic treatment,the protein expression levels of hemeoxygenase-1(HO-1)and nuclear factor erythroid 2-related factor 2 (Nrf2) in the brain tissue of BE group mice were increased.16S rDNA results showed that compared with the blank group (Con group),the abundances of Proteobacteria,Bacteroidetes,and Verrucomicrobia were significantly increased in PD mice,while the abundances of Actinobacteria, Patescibacteria, Campylobacterota, Cyanobacteria,and Deferribacteres were significantly decreased. Probiotic intervention corrected the above changes in the phylum-level intestinal microbiota of PD mice.The combination of E. faecium and B. subtilis has a significant therapeutic effect on MPTP-induced PD mice,and the mechanism may be related to antioxidant effects and the synergistic action of intestinal microbiota.
