中文摘要: 为获得适合酿造凤阳本土主栽葡萄品种‘巨峰’葡萄酒的专用酵母,选用李子、葡萄和石榴3种果皮,经过酵母浸出粉蛋白胨葡萄糖(yeast extract peptone dextrose,YPD)培养基富集、系列浓度稀释后涂布在5种分离培养基上,挑取菌落经过多次纯化后利用2,3,5-氯化三苯基四氮唑(triphenyltetrazolium chlorid,TTC)染色法和杜氏发酵法筛选高活力菌种,并进行‘巨峰’葡萄果酒发酵性能分析,最后对优异菌株进行分子鉴定。结果表明:不同果皮来源的酵母需要不同分离培养基,共筛选到起酵速度快、发酵液气味浓香的菌株60株;从中挑选11株酵母接入‘巨峰’葡萄汁获得浅红色葡萄酒,以商业酵母RW和SY为对照。11种葡萄酒的12项理化指标总体较好,除了Y11外,其他10种酒的柔和指数(suppleness index,IS)均大于6且高于2种商业酵母的发酵酒,尤其是Y4、Y7和Y10菌株发酵葡萄酒的IS超过10,后期可用作当地‘巨峰’葡萄生产桃红葡萄酒的优良菌株。经过内转录间隔区(internal transcribed spacer,ITS)序列比对,系统发育树结果显示Y4和Y10与汉逊德巴利酵母同源性分别为100%和99.84%,Y7与葡萄汁有孢汉逊酵母同源性为99.59%。
Abstract:To obtain specialized yeasts suitable for fermentation of Fengyang ′Kyoho′ grape wine,the yeasts from the plum,grape,and pomegranate peels were enriched by the yeast extract peptone dextrose(YPD) medium,diluted into serial concentrations,and coated on five separation media. The colonies were purified several times,and high activity yeasts were screened by means of the triphenyltetrazolium chlorid(TTC) chromogenic method and Duchenne tubular fermentation. The fermentation performance of ′Kyoho′ grape wine was analyzed,and excellent yeasts were identified by molecular technology. Results showed that yeasts from different peels required different isolation media,and a total of 60 strains with fast fermentation speed and strong flavor were selected. The 11 yeast strains were selected to inoculate into the ′Kyoho′ grape juice and obtain pale red grape wine,using commercial yeasts RW and SY as controls. The 12 physicochemical indicators of 11 wines were generally good. Except for Y11,the suppleness indexes (IS) of the other 10 wines were over 6 and higher than the two controls. In particular,the IS of wines fermented by Y4,Y7,and Y10 strains was more than 10,indicating these strains could be used as excellent choices for fermenting rose wine from Fengyang ′Kyoho′grapes. After internal transcribed spacer (ITS) sequence comparison,results of the phylogenetic tree showed that Y4 and Y10 were 100% and 99.84% homologous to Debaryomyces hansenii,respectively,and Y7 was 99.59% homologous to Hanseniaspora uvarum.
