中文摘要: 以富硒香菇为试材,采用纤维素酶协同超声辅助提取法,通过单因素试验结合响应面试验优化富硒香菇硒多糖的提取工艺,并对最优工艺下提取的香菇多糖(Lentinula edodes polysaccharides,LEP)和富硒香菇硒多糖(Sele‐nium rich Lentinula edodes polysaccharides,Se‐LEP)的体外抗氧化活性进行初步分析。结果表明:Se‐LEP 的最佳提取工艺条件为料液比1∶60(g/mL)、纤维素酶添加量7%、酶解时间2.0 h、超声时间50 min、pH4.0、酶解温度55 ℃、超声温度63 ℃、超声功率373 W,在此条件下Se‐LEP 的提取率为(32.09±0.11)%,硒含量为(5.32±0.45)mg/kg。最佳工艺下得到的Se‐LEP 对DPPH 自由基、ABTS+自由基和羟基自由基的清除率分别达到(53.46±0.21)%、(78.87±1.28)%和(49.19±0.21)%,清除能力均优于LEP。
Abstract:The cellulase assisted‐ultrasound extraction process of selenium‐rich Lentinus edodes polysaccha‐rides was optimized by single factor experiments combined with response surface methodology.The in vitro an‐tioxidant activities of L.edodes polysaccharides(LEP)and selenium‐rich L.edodes polysaccharides(Se‐LEP)extracted with the optimized process were evaluated.The results showed that the optimal extraction process for Se‐LEP was a solid‐to‐liquid ratio of 1∶60(g/mL),a cellulase addition amount of 7%,enzymatic hydrolysis for 2.0 h,ultrasound time of 50 min,pH4.0,enzymatic hydrolysis temperature of 55 ℃,ultrasound temperature of 63 ℃,and ultrasound power of 373 W. Under these conditions,the extraction rate and selenium content of Se‐LEP were(32.09±0.11)% and(5.32±0.45)mg/kg,respectively. The clearance rates of Se‐LEP extracted un‐der the optimal process against DPPH,ABTS+,and hydroxyl free radicals reached(53.46±0.21)%,(78.87±1.28)%,and(49.19±0.21)%,respectively,being higher than those of LEP.
