中文摘要: 该文建立鲜辣椒中辣椒素和二氢辣椒素的QuECHERS-液相色谱串联质谱检测方法。通过优化QuECHERS 前处理条件,提取鲜辣椒中的辣椒素和二氢辣椒素,用0.1% 甲酸乙腈溶液和0.1% 甲酸水溶液作为流动相,通过多反应监测(multiple reaction monitoring, MRM)、信息依赖性采集(information dependent acquisition,IDA)和增强子离子扫描(enhanced product ion scan,EPI)相结合的复合扫描模式进行检测分析,建立辣椒素和二氢辣椒素标准品EPI 二级谱库,通过保留时间、离子丰度比和谱库比对相结合,对目标化合物精准定性和定量检测。结果显示,辣椒素和二氢辣椒素在1.5~500.0 ng/mL 呈线性相关(R2=0.999),辣椒素和二氢辣椒素的方法定量限为0.087 mg/kg 和0.069 mg/kg,回收率为72.0%~110.3%,相对标准偏差低于8.0%。该检测方法检测时间短、灵敏度高,可有效排除假阳性,检测范围宽,可对不同辣度的辣椒中辣椒素和二氢辣椒素进行检测分析。

Abstract:A QuECHERS-liquid chromatography tandem mass spectrometry method for the detection of capsaicin and dihydrocapsaicin in fresh chilis was established. The pretreatment conditions of QuECHERS were optimized, and the capsaicin and dihydrocapsaicin in fresh chilis were extracted. 0.1% acetonitrile formate and 0.1% formic acid aqueous solution were used as mobile phases for detection and analysis by a combination of multiple reaction monitoring(MRM), information dependent acquisition (IDA), and enhanced product ion scanning (EPI). The secondary library of standard EPI of capsaicin and dihydrocapsaicin was established,and the target compounds were accurately qualitatively and quantitatively detected by combining retention time, ion abundance ratio, and library alignment. The results showed that capsaicin and dihydrocapsaicin were linear in the range of 1.5-500.0 ng/mL (correlation coefficient R2 = 0.999), and the limits of quantification of capsaicin and dihydrocapsaicin were 0.087 mg/kg and 0.069 mg/kg; the recovery rate was 72.0%-110.3%, and the relative standard deviation was within 8.0%. The detection method was featuring short detection time and high sensitivity. The false positive could be effectively excluded, and the detection range was wide. The capsaicin and dihydrocapsaicin in chilis with different pungency degrees could be detected and analyzed.
