中文摘要: 琼脂孔扩散法广谱抑菌试验时检测工作量大、效率低,培养皿分区法是在琼脂孔扩散法基础上进行改良的一种试验方法,用于提高广谱抑菌检测效率。该文采用琼脂孔扩散法与培养皿分区法分别对2 株产细菌素乳酸菌进行6 种指示菌抑菌性能检测,通过测定6 种指示菌抑菌圈直径(琼脂孔扩散法)与抑菌扇环环宽(培养皿分区法),比较分析两种方法广谱抑菌检测结果的一致性。采用琼脂孔扩散法与培养皿分区法在相同试验条件下,植物乳植杆菌(Lactiplantibacillus plantarum)CICC24194 培养上清液对6 种指示菌均具有抑制作用,对指示菌的抑菌能力均为单核细胞增生李斯特氏菌>金黄色葡萄球菌>志贺氏菌>克罗诺杆菌>沙门氏菌>大肠埃希氏菌。屎肠球菌(Enterococcus faecium)CICC24203 培养上清液对6 种指示菌均具有抑制作用,对指示菌的抑菌能力均为单核细胞增生李斯特氏菌>克罗诺杆菌>志贺氏菌>沙门氏菌>金黄色葡萄球菌>大肠埃希氏菌。培养皿分区法与琼脂孔扩散法广谱抑菌检测结果一致,培养皿分区法可在1 套培养皿中同时完成6 种指示菌的抑菌检测,试验结果准确、可靠、稳定,适用于广谱抑菌高效检测。
Abstract:The agar well diffusion assay for broad-spectrum bacteriostatic testing involves a large workload and is inefficient. The petri dish partitioning assay is an improved method based on the agar well diffusion assay,designed to enhance the efficiency of broad-spectrum bacteriostatic testing. In this study, the bacteriostatic properties of two bacteriocin-producing lactic acid bacteria against six indicator bacteria were tested using both the agar well diffusion assay and the petri dish partitioning assay. The consistency of the results from the two methods was compared and analyzed by measuring the inhibition zone diameters (agar well diffusion assay)and the inhibition annular sector widths (petri dish partitioning assay). Under the same test conditions, the culture supernatant of Lactiplantibacillus plantarum CICC24194 exhibited inhibitory effects against all six indicator bacteria. The bacteriostatic activity was as follows: Listeria monocytogenes > Staphylococcus aureus > Shigella > Cronobacter > Salmonella > Escherichia coli. The culture supernatant of Enterococcus faecium CICC24203 also inhibited all six indicator bacteria, with the bacteriostatic activity ordered as: L. monocytogenes > Cronobacter > Shigella > Salmonella > S. aureus > E. coli. The results of broad-spectrum bacteriostatic testing by the petri dish partitioning assay were consistent with those by the agar well diffusion assay. The petri dish partitioning assay allowed for the simultaneous testing of six indicator bacteria in one set of petri dishes,providing accurate, reliable, and stable results, making it suitable for efficient broad-spectrum bacteriostatic testing.
