半仿生-酶法提取葛根异黄酮类成分及其体外抗氧化活性

中文摘要: 采用半仿生-酶法提取葛根中异黄酮类成分,以基于葛根素、大豆苷和总异黄酮含量的综合评分为指标,结合单因素和正交试验,确定葛根异黄酮类成分最佳提取工艺,并对其体外抗氧化活性进行研究。结果表明,半仿生-酶法的最佳提取工艺条件为果胶酶添加量1.0%、酶解温度55 ℃、料液比1∶14 (g/mL),在此条件下,葛根素含量为63.287 mg/g,大豆苷含量为5.177 mg/g,总异黄酮含量为221.583 mg/g,综合评分为98.887;葛根异黄酮提取物对DPPH 自由基和ABTS+自由基的清除能力随其质量浓度的增加而提高,IC50 值分别为1.538 mg/mL 和0.748 6 mg/mL,优于30%乙醇超声辅助提取法。
Abstract:The semi-bionic enzyme method was used to extract isoflavonoids from Puerariae lobatae Radix. Using a comprehensive scoring system with the puerarin, daidzin, and total isoflavone content as indicators, this study determined the optimal extraction process for isoflavonoids from Puerariae lobatae Radix with single-factor and orthogonal experiments and explored their in vitro antioxidant activity. The findings revealed that the optimal extraction process conditions for the semi-bionic enzyme method included a pectinase dosage of 1.0%,an enzyme digestion temperature of 55 ℃, and a material-liquid ratio of 1∶14 (g/mL). Under these conditions, the puerarin content reached 63.287 mg/g, and the daidzin content stood at 5.177 mg/g. The total isoflavone content amounted to 221.583 mg/g, and the comprehensive score was 98.887 points. The scavenging ability of isoflavone extract from Puerariae lobatae Radix against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation free radicals increased with its concentration with IC50 values of 1.538 mg/mL and 0.748 6 mg/mL, respectively, which were superior to those of the isoflavone extract by the ultrasonic extraction method with 30% ethanol.
