间歇式微波辅助提取丹参总酚酸工艺优化及其酪氨酸酶抑制活性

中文摘要: 探讨间歇式微波辅助提取丹参总酚酸及其酪氨酸酶抑制活性。选取迷迭香酸、丹酚酸B 作为指标成分,以丹参总酚酸得率为响应值,通过单因素试验和响应面法考察液料比、提取时间、间歇比,获得间歇式微波辅助提取丹参总酚酸的最佳工艺,并利用酪氨酸酶抑制活性,评价间歇式微波辅助提取丹参总酚酸的技术优势。结果表明,丹参总酚酸的最佳提取工艺为液料比24.5∶1 (mL/g)、提取时间15 min、间歇比1∶1。在最佳提取条件下,丹参总酚酸得率为7.84%,其抑制酪氨酸酶活性半数抑制浓度(median inhibition concentration,IC50)为11.6 μg/mL。分子对接结果显示,迷迭香酸、丹酚酸B 与酪氨酸酶的结合能分别为-8.2、-7.5 kcal/mol(1 kcal/mol=4.184 kJ/mol),与铜离子周围的氨基酸残基(如GLY-389、THR-31、SER-394)相互作用形成稳定的复合物,从而抑制酶的活性。
Abstract:The intermittent microwave-assisted extraction of total phenolic acids from?Salvia?miltiorrhiza?and their inhibitory activity on tyrosinase were investigated. Rosmarinic acid and salvianolic acid B were selected as the index components, and the yield of total phenolic acids from?Salvia?miltiorrhiza?was taken as the response value. The optimal process for intermittent microwave-assisted extraction of total phenolic acids from?Salvia?miltiorrhiza?was obtained by investigating liquid-material ratio, extraction time, and intermittent ratio through the single factor experiments and response surface methods. Meanwhile, the technical advantages of intermittent microwave-assisted extraction of total phenolic acids from?Salvia?miltiorrhiza?were evaluated by using their inhibitory activity on tyrosinase. The results show that the optimal parameters for extracting total phenolic acids from?Salvia?miltiorrhiza?were as follows: liquid-solid ratio of 24.5∶1(mL/g), extraction time of 15.0 min, and intermittent ratio of 1∶1. Under the optimum extraction condition, the yield of total phenolic acids from?Salvia?miltiorrhiza?was 7.84%, and the inhibitory activity on tyrosinase IC50?value was 11.6 μg/mL.The molecular docking results showed that the binding energy of rosmarinic acid and salvianolic acid B with tyrosinase were -8.2, -7.5 kcal/mol(1 kcal/mol=4.184 kJ/mol), respectively. It interacted with amino acid residues (such as GLY-389, THR-31, and SER-394) around copper ions to form stable complexes and thereby inhibited the enzyme activity.
