大米谷蛋白/表面活性剂纳米颗粒的构建及其对姜黄素的递送功效

中文摘要: 姜黄素具有抗炎、抗氧化等功效,但水溶性和物理化学稳定性较差限制了姜黄素广泛应用。为改善姜黄素的水溶性、稳定性及生物利用度,该文利用pH 驱动法成功构建一种稳定的大米谷蛋白和表面活性剂递送体系,同时研究表面活性剂的种类(阴离子表面活性剂鼠李糖脂和两性表面活性剂卵磷脂)和大米谷蛋白与表面活性剂质量比对复合物功能属性的影响。结果表明,大米谷蛋白与鼠李糖脂、卵磷脂的最佳质量比分别为5∶1 和2∶1。在此质量比条件下,复合物具有最小的粒径,分别为(138.30±1.77) nm 和(199.93±1.85) nm 以及最高的包埋率,分别为(93.56±0.41)%和(66.85±0.07)%。扫描电子显微镜观察结果表明,加入姜黄素及表面活性剂后,复合物的形态更均匀。荧光光谱显示,加入姜黄素及表面活性剂后复合物荧光强度降低。此外,由阴离子表面活性剂鼠李糖脂构建的纳米递送系统表现出更高的光稳定性、热稳定性以及体外消化过程中较高的胃酸稳定性和可控的姜黄素释放率。综上,由大米谷蛋白和鼠李糖脂构建的纳米递送系统可以作为姜黄素良好的载体,从而提高姜黄素的生物活性。
Abstract:Curcumin has benefits such as anti-inflammation and antioxidant properties. However, its poor water solubility and physicochemical stability limit its widespread application. To improve the water solubility,stability, and bioavailability of curcumin, a stable delivery system of rice glutelin and surfactant was constructed by the pH-driven method. Additionally, the effects of surfactant types (anionic surfactant rhamnolipid and amphoteric surfactant lecithin) and mass ratios of rice glutelin to surfactant on the functional properties of the complexes were studied. The results showed that the optimal mass ratios of rice glutelin to rhamnolipid and lecithin were 5∶1 and 2∶1, respectively. At these ratios, the smallest particle sizes were (138.30±1.77) and(199.93±1.85) nm, respectively, along with the highest encapsulation rates of (93.56±0.41)% and (66.85±0.07)%, respectively. Scanning electron microscopy (SEM) observations indicated that the addition of curcumin and surfactant resulted in a more uniform morphology of the complexes. Fluorescence spectroscopy demonstrated that the fluorescence intensity decreased after the addition of curcumin and surfactant. In addition,the nano-delivery system constructed with the anionic surfactant rhamnolipid exhibited superior photostability,thermal stability, gastric acid stability, and a controllable curcumin release rate during in vitro digestion. In conclusion, the nano-delivery system based on rice glutelin and rhamnolipid could serve as a good carrier for curcumin, thus improving the biological activity of curcumin.
