黄芪茎叶总黄酮的超声辅助双水相提取工艺优化及其抗氧化活性

中文摘要: 该研究采用超声辅助乙醇-硫酸铵双水相体系提取黄芪茎叶总黄酮,以总黄酮提取量为指标,通过单因素试验结合响应面试验优化提取工艺,并对所得总黄酮提取物的抗氧化活性进行分析。结果表明,超声辅助双水相提取黄芪茎叶总黄酮的最佳工艺条件为乙醇体积分数60%、硫酸铵质量浓度0.20 g/mL、液料比31∶1(mL/g)、超声时间50 min、超声温度46 ℃、超声功率480 W,此条件下黄芪茎叶总黄酮提取量为15.008 mg/g。所制备的黄芪茎叶总黄酮提取物表现出良好的抗氧化活性,其清除DPPH·、ABTS+·和·OH 的IC50 值分别为0.933、0.691、0.965 mg/mL,2.0 mg/mL 总黄酮提取物溶液的铁离子还原能力(ferric ion reducing antioxidant power,FRAP)为1.384 mmol/L,是同浓度阳性对照VC 铁离子还原能力的83.47%。

Abstract:Total flavonoids of Astragalus membranaceus stems and leaves were obtained by ultrasonic-assisted extraction in an ethanol-ammonium sulfate aqueous two-phase system.With the extraction amount of total flavonoids as an index,the extraction process was optimized by combining the single-factor experiments with response surface analysis,and the antioxidant activity of the total flavonoids was also investigated. The results showed that the optimal conditions for ultrasonic-assisted aqueous two-phase extraction of total flavonoids from Astragalus membranaceus stems and leaves were as follows:ethanol volume fraction of 60%,ammonium sulfate concentration of 0.20 g/mL,liquid-solid ratio of 31∶1(mL/g),ultrasonic time of 50 min,ultrasonic temperature of 46 ℃,and ultrasonic power of 480 W. Under these conditions,the amount of total flavonoids was 15.008 mg/g. The prepared total flavonoid extracts from Astragalus membranaceus stems and leaves exhibited good antioxidant activity. The IC50 values for scavenging DPPH·,ABTS+·,and ·OH were 0.933,0.691,and 0.965 mg/mL,respectively.When the concentration of the total flavonoids was 2.0 mg/mL,its ferric ion reducing antioxidant power(FRAP)value was 1.384 mmol/L,which was 83.47%of that of the positive control VC at the same concentration.
