中文摘要: 为探究叶黄素酯-虾青素复合物对蓝光发光二极管(light-emitting diodes,LED)诱导的视网膜色素上皮细胞ARPE-19 及SD 大鼠视网膜组织损伤的保护作用,采用不同浓度的叶黄素酯-虾青素复合物预处理细胞6 h,实验分为6 组:对照组、损伤组、参考组、低剂量组、中剂量组、高剂量组,蓝光LED 下光氧化损伤诱导12 h。采用噻唑蓝法测定细胞活力,检测乳酸脱氢酶释放率分析细胞损伤程度,2′,7′-二氯二氢荧光素二乙酸酯荧光探针标记检测细胞内活性氧(reactive oxygen species,ROS)水平。利用蓝光损伤装置建立大鼠视网膜损伤模型,在光强度为(5 500±100)lux 条件下暴露10 h 后,暗室(0 lux)饲养24 h。苏木精-伊红染色组织切片可观察SD 大鼠视网膜形态学变化,测定组织中的氧化应激相关指标。结果显示:叶黄素酯-虾青素复合物预处理可有效防止蓝光诱导的ARPE-19 细胞活力降低,防止细胞损伤;显著降低细胞内ROS 水平。同时,中剂量组和高剂量组显著提升超氧化物歧化酶、谷胱甘肽过氧化物酶活力,降低组织中的丙二醛含量,改善SD 大鼠视网膜组织免受蓝光诱导的细胞氧化应激和组织病理损伤。
Abstract:The protective effects of lutein ester-astaxanthin complex on the retinal pigment epithelial cell line ARPE-19 and the retinal tissue of Sprague-Dawley(SD)rats exposed to blue light-emitting diodes(LED)were studied.The cells were classified into control,damage,reference,low-,medium-,and high-dose lutein ester-astaxanthin complex groups and pretreated for 6 h.The photo-oxidative damage was induced by blue LED for 12 h. The thiazolyl blue assay was employed to determine the viability of cells,and the degree of damage was evaluated by the lactate dehydrogenase release rate. The fluorescent probe 2′,7′-dichlorodihydrofluorescein diacetate was used to measure the intracellular reactive oxygen species(ROS)level. A blue light device was used to establish a rat retinal damage model. Specifically,the rats were exposed to blue light at(5 500±100)lux for 10 h and then kept in a dark room(0 lux)for 24 h. Hematoxylin-eosin staining of tissue sections was adopted to observe the morphological changes in the retinae of SD rats,and the oxidative stress indicators in the retinal tissue.The results showed that pretreatment with lutein ester-astaxanthin complex effectively prevented the blue light-induced reduction in ARPE-19 cell viability,protected the cells from damage,and lowered the intracellular ROS level.Meanwhile,the medium-and high-dose lutein ester-astaxanthin complex significantly enhanced the superoxide dismutase and glutathione peroxidase activities and lowered the malondialdehyde content in the tissue,thus protecting the retinae of SD rats from blue light-induced cellular oxidative stress and histopathological damage.
